原核表現H6N1亞型家禽流行性感冒病毒血球凝集素在雞隻的免疫反應

Immunogenicity of Prokaryotic Expression H6N1 Avian Influenza Virus Hemagglutinin in Chickens

摘要


在臺灣H6N1亞型的家禽流行性感冒病毒 (Avian influenza virus, AIV,簡稱禽流感病毒)已存在43年,造成業者的損失,開發一有效的原核表現蛋白質疫苗,防止本病的發生為本研究的目的。本研究針對H6N1病毒株的血球凝集素(hemagglutinin, HA)基因序列上HA_1誘導產生中和抗體區域,進行選殖,並構築於原核表現系統載體pGEX-5X-1上進行蛋白質的表現,以發展次單位疫苗,將表現出的蛋白質進行純化後以皮下注射方式免疫接種於4週齡雞隻,2週後進行補強注射,在免疫及補強注射後2週採血,以血球凝集抑制(Hemagglutination inhibition, HI)試驗檢測H6N1亞型HI抗體力價,評估其免疫性。結果免疫雞隻抗H6N1 HI抗體達2^(6.2),顯示AIV H6N1亞型HA蛋白質對雞隻的免疫效果良好,可做為控制H6N1禽流感的次單位疫苗。

The H6N1 low pathogenic avian influenza virus has been present in Taiwan for 43 years and has caused substantial economic losses in the poultry industry. To develop a prokaryotic expression hemagglutinin (HA) protein for controlling this disease was the purpose of this stundy. The HA1 gene relating to epitopes was cloned and expressed in pGEX-5X-1. The expression protein was purified and injected subcutaneously into 4-week-old chickens and then boosted 2 weeks after. Blood was collected at two weeks after the second vaccination for a hemagglutination-inhibition (HI) test. The results showed that this expression protein could induce HI antibody titer of 2^(6.2) in chickens. In conclusion, this expression protein shows possibilities for use as a subunit vaccine for controlling the H6N1 avian influenza infection in chickens.
