以黏粒修飾電極法探討地衣芽孢桿菌之電化學反應

Electrochemical Probing of Bacillus Licheniformis on Clay-Modified Electrodes

摘要


黏粒修飾電極可用於重金屬、農藥及生化物質等的偵測,由於黏粒層的電子傳導特性也常被用於電化學分析、光催化性與生化感測器等用途。以較具有電化學活性的Fe (CN)_6^(3-)及Ru(NH_3)_6^(3+)作為探針化合物,利用黏粒修飾電極搭配循環伏安法來探討地衣芽孢桿菌表面的電化學特性。試驗結果得知,PFl-1與菌混合液修飾電極對Fe (CN)_6^(3-)偵測到的氧化還原波峰電流量隨第一圈到第五圈逐漸下降,且氧化波峰形狀較為尖銳,由電極表面的掃描式電子顯微鏡圖可明顯看到地衣芽孢桿菌的形狀。SWy-1與菌混合修飾電極對Fe (CN)_6^(3+)偵測到的還原波峰形狀較為駝形,其還原波峰電流量隨掃描第一圈到第五圈呈現有規則性逐漸減少的現象。SWy-1與菌混合修飾電極對Ru (NH_3)_6^(3+)測到的氧化還原波峰電流量比SWy-1修飾電極所測到的小很多。由黏粒修飾電極的電化學分析結果顯示以Fe (CN)_6^(3-)作為探針分子較Ru(NH_3)_6^(3+)能顯現地衣芽孢桿菌在電化學反應上的特色。

Clay-modified electrodes have been applied for the determination of heavy metals, pesticides and biochemical compounds. In addition to electroanalysis, there are also extensive studies on photocatalysis and biosensors due to the good conductivity of the clay layer. In this study, two probes Fe(CN)_6^(3-) and Ru(NH3)_6^(3+) were used to study the electrochemical reaction of Bacillus licheniformis at clay modified electrodes. The redox peak currents of Fe(CN)_6^(3-) at the electrode modified with added PFl-1 bacteria were found to decrease from first to fifth cycle with the shape of oxidative peak being sharper than other modified electrodes. Field emission SEM clearly showed the rob form of B. licheniformis on the electrode modified with added PFl-1 bacteria. At the SWy-1 added bacteria-modified electrode, the reductive peak currents of Fe(CN)_6^(3-) were found to regularly decrease from first to fifth cycle. In contrast, the decrease was much less for Ru(NH3)_6^(3+) at the electrode modified with added SWy-1 bacteria. Overall, Fe(CN)_6^(3-) appears to be a favorable probe for the electrochemical reaction of B. licheniformis on clay-modified electrodes.
