Osteoimmunology: The Relationship between T Cells and Alveolar Bone Resorption

摘要


牙周病是由口腔內細菌所引發的發炎性疾病,當牙周組織受到細菌入侵之後,自然地,體內會自動啟動免疫反應。近年來,發現免疫系統裡的T細胞可以直接地或間接地使得RANKL增多,RANKL是啟動噬骨細胞生成的主要物質,所以T細胞和骨吸收之間的關係也成了熱門的研究主題,有學者甚至提出了osteoimmunology這個新名詞,來突顯出免疫系統和骨頭重塑間的關聯及重要性,這是一門新興的學問。本文將探討T細胞主導牙周病骨吸收的過程,從T細胞是在何種條件下產生RANKL、T細胞如何直接及間接地使RANKL增多,以及T細胞如何干涉RANKL在噬骨前驅細胞內的訊息傳導,讓大家了解T細胞在骨質喪失中所扮演的巧妙角色。

Periodontal diseases are inflammatory conditions which are induced by bacteria in the oral cavity. After bacterial invasion into the periodontal tissue, an immune response is initiated within the human body. Recently, it was discovered that T lymphocytes play an important role in adjusting, both directly and indirectly, the amount of receptor activator of nuclear factor KB ligand (RANKL) in the process of bone resorption. RANKL is the critical messenger that signals the formation of osteoclasts. Therefore, the relationship between T cells and bone resorption is a research topic worthy of further and more-thorough investigation. Some scholars even use the new term ”osteoimmunology” to describe the importance of the relationship between the immune system and bone remodeling. In this article, we discuss T cells' influential role in alveolar bone resorption. More specifically, we focused on 2 aspects: (1) T cells' mode of action in inducing RANKL production via both direct and indirect pathways and (2) their ability to interfere with RANKL signals in osteoclast precursors.
