RUNX3是一種新世代的抑癌基因。它被發現與胃的粕膜細胞增生有密切的關係。如果 RuNx3 抑癌基因發生突變,在動物實驗中可以發現胃黏膜細胞有異常增生的現象。RUNX3抑癌基因發生突變,主要是在其 Promotor 位置發 Hypermethylation 。我們回朔性的將11位殘冑癌的病患的手術標本做 DNA 分析,發現其中確實有 RUNX3 Promotor Hypermethy-lation 的現象。
RUNX3 (PEBP2aIphaC/CBFA3/AML2) is a transcription factor downstream of the TGF p signaling pathway. RUNX3 knocked-out mice showed hyperproliferation and hyperplasia in gastric epithelium. Lack of RUNX 3 expression, partly due to promoter hypermethylation, had been associated with the human gastric cancers. This study is aimed to evaluate the relationship between RUNX3 expression and human remnant gastric cancer. The methylation status of RUNX 3 promoter in human remnant gastric cancer was investigated by methylation-specific restriction enzyme digestion and PCR amplification. We retrospectively reviewed the surgically removed specimen of eleven cases of remnant gastric cancer. Hypermethylation of RUNX 3 in its promoter region was found in the tumor tissues of all eleven cases. Further studies are required to clarify the direct relationship between RUNX3 and remnant gastric cancer.
