The Effect of Prostaglandin E1 on Vasoconstriction Induced By Extravasating Blood

The hind limbs of adult rats were nearly completely amputated at mid-thigh level leaving the femoral artery and vein intact. The probe of a Laser Doppler Flowmeter was placed on the heel pad which reflected the severity of femoral vasospasm when extravasting blood from another rat was applied to the vessels. The perfusion of that limb was reduced to 40% of its original state after the clotting blood contacted the femoral vessels. This vasoconstriction effect was prevented or rapidly relieved by intra-arterial infusion of prostaglandin E1.
