番茄殘渣多元酚對癌化細胞增生及第二型環氧合酶活性抑制之研究

Study on the Proliferation and Cyclooxygenase II Activity of Carcinoma Cells by Tomato Residue Polyphenols

摘要


番茄為具有保健功效之天然蔬果,攝取之來源以其加工品為主(如番茄汁、醬等),但番茄加工時產生許多殘渣,本研究室已發現番茄殘渣含有高量之多元酚,且這些多元酚擁有極佳之抗氧化性。口腔癌及攝護腺癌為台灣主要的癌症,為瞭解番茄殘渣多元酚對此二種癌症是否具有預防功效,本研究擬以口腔鱗狀上皮癌細胞(KB cell)及攝護腺癌細胞(PC-3 cell)為模式系統,探討經不同乾燥處理之番茄殘渣的多元酚粗萃物,是否能有效地抑制癌細胞之增生以及第二型環氧合酶(cyclooxygenase-2;COX-2)之表現。結果顯示,番茄殘渣多元酚粗萃物能顯著抑制口腔鱗狀上皮癌細胞及攝護腺癌細胞的增生,且抑制作用與劑量成正相關,其中經高溫乾燥處理(140℃)之番茄殘渣的多元酚粗萃物具有最佳之效果。同樣地,番茄殘渣中的多元酚粗萃物對COX-2之表現亦有顯著的抑制效果,且抑制能力與劑量成正相關,仍然是以高溫乾燥處理者(140℃)效果最好。由本研究可知,原欲丟棄之番茄殘渣中,可能含有癌症預防功效之多元酚,未來有必要作進一步利用與探討。

Tomato is a fresh functional vegetable. The main intake of tomato occurs through related processed products(e. g., tomato juice and ketchup). Most tomato processing leads to a great amount of residues. This laboratory found that tomato residues contain high levels of polyphenols, which show excellent antioxidant activities. Prostate and oral cancers are two important cancers in Taiwan. This study focused on the prevention of both oral and prostate cancers by tomato residue polyphenols. The effect of tomato residue polyphenols on the proliferation and cyclooxygenase II activities of both oral squmaous(KB)and prostate(PC-3)carcinoma cells is discussed in this study. Results showed that tomato residue polyphenols effectively inhibited the proliferation and cyclooxygenase II activities of both KB and PC cells in a dose-dependent manner. The polyphenols obtained from tomato residues by high temperature drying (140℃) showed the best effect. According to this study, tomato residue polyphenols can probably be helpful in cancer prevention. Further studies are required in the future.
