Production of PR-imine, PR-acid, and PR-amide Relative to the Metabolism of PR Toxin by Penicillium Roqueforti

藍酪黴菌產生與 PR toxin 相關之代謝產物PR-imine、PR-acid 及 PR-amide 之研究

摘要


本研究利用高效能液態層析儀測定藍酪黴菌產生藍酪黴毒素 (PR toxin) 及其代謝產物 PR-imine、PRacid及 PR-amide 之產量。在供試四株藍酪黴菌中只有二菌株 (ATCC 48779 及 ATCC48936) 具有產生此四種次級代謝產物之能力,而且此些化合物主要存在培養基中,菌體中之產量很少;產生此些次級代謝產物之最適 pH 值為 4.0。在菌體生長期間顯示伴隨藍酪黴毒素產量之下降,PR-imine、PR-acid及 PR-amide 化合物隨之產生;此外,此四個化合物之結構很相似。從其個別產生之時間順序消長關係及化學結構之相關性資料,我們提出有關藍酪黴毒素代謝途徑之假說:藍酪黴毒素部分代謝成為PR-imine,另一途徑則代謝為 PR-acid 然後可能再代謝為 PR-amide。

The production of PR toxin and the closely related metabolites, PR-imine, PR-acid, and PR-amide by four Penicillium roqueforti isolates was analyzed by high-performance liquid chromatography (HPLC). Only two isolates (ATCC 48779 and ATCC 48936) were able to produce all four compounds. Moreover, the yields of these compounds were lower in the mycelium than in the YESC culture medium. The optimal pH for the production of these compounds was around pH 4.0. In a time-course study, the appearance of PR-imine, PRacid, and PR-amide coincided with the disappearance of PR toxin. Besides, the chemical structures of the four compounds are closely related. Based on these data we propose that there are two metabolic pathways for PR toxin. It is metabolized into either PR-imine or PR-acid. The latter might be further metabolized into PRamide.
