水稻紋枯病菌之雙股核醣核酸的研究

Study on the Double-Stranded RNAs in Different Isolates of R. solani AG-1 IA

摘要


由供試水稻紋枯病菌 (Rhizoctonia solani AG-1 IA) 菌株的菌絲與菌核分離的 dsRNAs 片段可分為 2.0kb、2.5 kb、3.0 kb 及 14.0 kb,其中 2.0 kb 片段在 Rs1E-2、Rs1F-1、Rs1H、Rs1M-3、Rs1N-2、Rh-Ta、PR-06、Rh-cabbage、Rh-Tsau (3) 等菌株中皆有發現。再從供試菌株進行水稻接種試驗後所得到的新分離株,進行 dsRNA 的抽取,發現 dsRNA 存在的情形與原來未接種前的供試菌株中所具有的dsRNA 片段大小是一致的,此外分別在健康與接種 Rs1M-3 的罹病水稻中抽取 dsRNA,都沒有任何dsRNA 的存在。由 Rs1M-3 與 PR-06 的細胞殘體、細胞核、細胞質、粒腺體中抽取 dsRNA,只有在細胞殘體與細胞質中發現有 dsRNAs 存在。在四次繼代培養的 Rs17 與 Rs1M-3 菌株中仍能發現dsRNAs 可以穩定存在於菌株中。因此,水稻紋枯病菌菌株具有雙股核醣核酸,其存在意義有待進一步探討。

Four double-stranded RNA elements (dsRNA) were isolated from native isolate of Rhizoctonia solani AG-1 IA in Taiwan. The sizes of dsRNA are 2 kb, 2.5 kb, 3 kb, and 14 kb, in which 2 kb dsRNA were found in the isolates of Rs1E-2, Rs1F-1, Rs1H, Rs1M-3, Rs1N-2, Rh-Ta, PR-06, Rh-cabbage, and Rh-Tasu (3). The dsRNA elements in new isolates collected from plants infected tested isolates were the same with tested isolates. The dsRNA elements were present in the cell debris and microsomal fractions of isolates Rs1M-3 and PR-06. The dsRNA can exist in cells of Rs17 and Rs1M-3 isolates via serially transferred up to four generations on PDA. From this study, we suggest the dsRNA are commonly associated with in R. solani AG-1 IA.
