Polymorphism of ITS Sequences of Nuclear Ribosomal DNA in Metasequoia Glyptostroboides

The internal transcribed spacers (ITS) and the 5.8S coding region of nuclear ribosomal DNA of Metasequoia glyptostroboides were amplified and sequenced using samples collected from its native population in south-central China and cultivated tree in Nanjing City. For the native Metasequoia population and cultivated tree, there was no variation in length of ITS region with ITS1, 702 bp; ITS2, 217 bp and 5.8S, 162 bp, and no nucleotide substitutions were found of all samples sequenced, but there were polymorphic sites (different nucleotides at one site) detected in all samples, the number of polymorphic sites is variable in different samples, ranging from 14 to 56. Phylogenetic analysis based on ITS region sequences with PHYLIP showed that an isolated tree in Hunan Province is more distant from other trees and the other trees are closely related. This result is consistent with the new leaf cuticle micromorphology studies of native Metasequoia population.
