在逆微胞系統中以脂解酵素製造建構脂肪之研究

A Study of the Production of Structured Lipids by Lipase in a Reverse Micellar System

本研究以三油酸甘油酯(triolein)及辛酸(caprylic acid)為反應基質,利用固定化脂解酵素IM77在逆微胞系統中催化合成建構脂肪(structured lipids),並使用反應曲面法及三階層四變數實驗設計以評估反應變數,包括反應時間(4-12h)、反應溫度(25-65ºC)、添加水量(0-10%)及界面活性劑濃度(0-400mM)對建構脂肪之莫耳轉換率的影響。研究結果顯示,高溫短時間或低溫長時間的條件組合皆可增加建構脂肪之莫耳轉換率;添加水量及提高界面活性劑濃度皆會降低其莫耳轉換率。經過多重迴歸統計分析後,得到最適合成條件為:反應時間12h、反應溫度 50°C、不添加水及界面活性劑,預測理論上,最適的莫耳轉換率為85%。

The enzymatic transesterification of 25 mM triolein and 100 mM caprylic acid (C8) to structured lipids (SL) with an immobilized lipozyme IM77 in a reverse micellar system was investigated in this study. Response surface methodology (RSM) and a three-level, four-factor experimental design were used to evaluate the effects of synthesis variables, including reaction time (4-12h), temperature (25-65°C), added water content (0-10%), and surfactant concentration (0-400mM) on the percent molar conversion of SL by transesterification. The result showed that the reaction at a high temperature and short time or a low temperature and long time increased the yield of SL. Added water content and dioctyl sulfosuccinate (AOT) concentration decreased the yield of SL. Based on the contour plots, the optimum synthesis conditions were: reaction time, 12h; reaction temperature, 50°C; added water, 0%; and AOT concentration, 0mM. The percent molar conversion of the optimum point was 85%.
