摘要


紅龍果(Hylocereus undatus Britt. & Rose)為仙人掌科多年生攀緣性肉質植物,所結果實汁多味美、營養豐富,是近年來相當受重視的新興熱帶栽培果樹。本研究於1999年在新竹縣關西鎮發現紅龍果三角莖上有黃綠參雜之斑駁病徵,初步 子顯徵鏡觀察,發現有絲狀病毒粒子存在。為了確認病毒種類,先以汁液接種法接種至紅藜,並經三次單斑分離後得到一病毒分離株。進一步進行指示植物接種、病毒粒子純化及雙向免疫擴散反應之分析結果顯示,引發紅龍果產生斑駁病徵之病原應為仙人掌病毒X(Cactus virus X, CVX)之一分離株,其屬於Potexvirus屬,可感染多種仙人掌科植物。由於紅龍果主要以扦插繁殖,病毒很容易藉此述徑傳播開來,本研究針對自紅龍果所分離之CVX-Hu製備抗血清,並建立夾層酵素聯結免疫分析技術,以其具有操作簡易快速及可同時檢測大量樣本之優點,將是進行紅龍果CVX檢測及無毒扦插繁殖體篩選最適合之技術。

Hylocereus undatus Britt. & Rose belongs to Cactaceae. It produces delicious fruits and has recently become a fruit crop of economic importance in Taiwan. In 1999, H. undatus grown in Kuan Shi (Hsin Chu, Taiwan) was found to display systemic mild mottlings on the trigonal stems. Preliminary analysis by electron microscopy of the sap prepared from diseased plants revealed the presence of flexuous rod-shaped virus particles. A viral strain was isolated from diseased H. undatus and established by three successive single lesion transfers on leaves of C. amaranticolor. Host range studies and serological analysis of purified virus indicate that the pathogen causing mottle disease on Hylocereus undatus is a strain of Cactue virus X (CVX). A procedure using antisera prepared against virus was established for detection of CVX by DAS-ELISA. Since H. undatus reproduces asexually, rapid and specific detection of the virus by DAS-ELISA will help to implement a sound control of the disease caused by CVX in H. undatus.
