Rapid Identification of Microorganism: Application of Flow Cytometry/Fluorescence Indicator

摘要


許多病原菌,包括結核菌,靠傳統的培養法需花很多時間,無法達成我們期望的快速、高敏感性及高專一性的地步。流式細胞儀(或稱螢光啟動細胞分離儀)配合螢光標記物(或稱螢光染劑)的使用,是目前衆多發展方法中引人注目的一種。它有許多好處,包括無放射性、可用完整的細胞或細菌來偵測、價格便宜、操作時間短、被紫外線激發螢光會大量釋出、數種螢光物質放在一起彼此不會幹擾、以及可正確地算出病原菌數或抗菌藥物濃度。加用專一抗體結合螢光再配合流式細胞儀,已可快速地檢驗出結核菌,不久這種儀器將可上市,和polymerase chain reaction衍生技術所登展出來的儀器一爭長短。

Many pathogens, including mycobacterium tuberculosis, need a long time to be identified by traditional culture method. This is beyond our expectation of rapidity, high sensitivity and high specificity. Flow cytometry/fluorescence indicator is one of the newly developments which attract our attention. This method has many advantages, including: no radiation, dealing with intact cells or pathogens, low cost, rapidity, high quantum yield after absorbing ultraviolet light, no interference of fluorescence indicators when they are mixed up, and correct estimation of the number of pathogens or the concentration of antibiotics. When specific antibody is linked to fluorescence indicator and is used along with flow cytometry, it has been demonstrated to rapidly detect mycobacterium tuberculosis. In the near future, this kind of machines may come into the market to compete with the other designs made of polymerase chain reaction derived techniques for the rapid identification of slow-growing pathogens.
