In a current study, chicken leg bone protein was hydrolyzed by Alcalase and investigated its angiotensin I converting enzyme (ACE) inhibitory activity. Hydrolysate with the strongest ACE inhibitory activity was selected as potent ACE-I and administrated orally in spontaneously hypertensive rats (SHR) for eight weeks to study changes in systolic blood pressure (SBP) and diastolic blood pressure (DBP). The results showed that chicken leg bone protein hydrolyzed by Alcalase for 4 hrs (A4H) exhibited the highest ACE inhibitory activity (IC50=0.545 mg/mL). After oral administration, SHRs treated with A4H by 50 mg/kg bw/day for eight weeks had lower SBP and DBP than control SHRs. Moreover, there was no significant difference in average daily feed intake (ADFI) and average daily drink intake (ADDI) between SHRs treated with A4H and control group. The present results suggest that A4H which derived form chicken leg bone could be utilized for developing ACE inhibitor as a potential ingredient of functional food for antihypertension.
