Fractionation of Lipolysis-stimulating Peptides Derived from Neutrase-soy Protein Hydrolysate Using Ultrafiltration Membrane

The aim of this study was to explore the lipolysis-stimulating activity of isolated soy protein (ISP) hydrolysate using 3T3-L1 adipocytes. Extracellular glycerol release (GR) and intracellular triglyceride residual (TR) were employed as markers for lipolysis in cells. The higher GR or the lower TR represents better lipolysis-stimulating activity. ISP was hydrolyzed with Neutrase for 4 h to obtain the hydrolysate NH4h, which showed lipolysis-stimulating activity in adipocytes at 400-800 ppm levels. The highest GR was observed at 400 ppm. GR increased from 79.03 to 109.24 nmol/mL, and TR decreased from 3.06 to 2.85 μmol/mg protein when compared to control. Both markers showed the efficacy of NH4h to stimulate lipolysis in the cells. The 10 kDa concentrate obtained from the sequential fractionation of NH4h with 30, 10 and 1 kDa molecular weight cut-off membranes (MWCO) resulted in further enhancement of its lipolysis-stimulating activity in the cells. The GR increased from 108.86 to 114.88 nmol/mL, and TR decreased from 2.85 to 2.35 μmol/mg protein at the 400 ppm level (p<0.05) when compared to the NH4h-added sample.
