添加外源性一氧化氮、細胞激素與脂多醣體對吳郭魚頭腎來源巨噬細胞發生apoptosis之影響

The Effects of Nitric Oxide Donors, Cytokines, and Lipopolysaccharide on Apoptosis of the Tilapia Head Kidney-derived Macrophages

摘要


本研究之目的在於探討一氧化氮(nitric oxide; NO)、細胞激素與脂多醣體(lipopolysacharide; LPS)對吳郭魚頭腎巨噬細胞免疫功能之影響,並嘗試以流式細胞儀(flow cytometer)來分析NO對頭腎巨噬細胞的影響。實驗結果發現頭腎巨噬細胞如分別以干擾素-γ(interferon-γ; IFN-γ)和LPS、腫瘤necrosis因子-α(tumor necrosis factor-α; TNF-α)及LPS與IFN-γ作用24小時後,均能誘發頭腎巨噬細胞發生apoptosis,同時發現IFN-γ亦可誘發頭腎巨噬細胞發生necrosis。然而,當添加低濃度的NO donors時,結果發現會抑制經IFN-γ與LPS作用過之頭腎巨噬細胞發生apoptosis,同時亦發現介白素-4(interleukin-4; IL-4)和介白素-10(interleukin-10; IL-10)均會抑制頭腎巨噬細胞發生apoptosis。另外,實驗結果顯示頭腎巨噬細胞經高濃度之NO donors誘導24小時後,頭腎巨噬細胞會發生apoptosis,並且其發生之比例會隨著NO donors的濃度的提高而增加。

The aims of this investigation were to study the effects of nitric oxide donors, cytokines, and lipopolysaccharide (LPS) on the non-specific immune responses of the tilapia (Oreochromis hybrids) head kidney-derived macrophages by using flow cytometry. Our results showed that tilapia head kidney-derived macrophages treated with IFN-γ, TNF-α, and IFN-γ plus LPS for 24 h exhibited a very high percentage of apoptotic cells. At the same time, IFN-γ also induced apoptosis in tilapia head kidney-derived macrophages. However, low doses of nitric oxide donors were able to inhibit apoptosis in the cells. The percentage of apoptosis of the tilapia macrophages decreased with addition of IL-4 and IL-10. On the other hand, tilapia head kidney-derived macrophages treated with high doses of nitric oxide donors for 24 h expressed a very high percentage of apoptosis. Simultaneously, the effect seemed to be dose-dependent.
