銀耳凍晶微脂粒之製備及有效性評估

Preparation of "Tremella Fuciformis" Lyophilized Liposome and Evaluation of Its Efficacy

摘要


本研究將銀耳萃取液以薄膜/乳化均質/超音波震盪法製備成微脂粒後,以冷凍乾燥法製成凍晶。實驗除進行微脂粒粒徑測量外,並以三種凍晶還原液進行皮膚保濕效果及彈性評估。實驗結果發現,以薄膜/乳化均質法製備A、B配方之微脂粒,其粒徑分別為1563 ± 164nm與1503 ± 415nm,而薄膜/乳化均質/超音波震盪法之粒徑分別為262.2 ± 4.90nm與186.6 ± 9.53nm。皮膚角質層水分含量測試B、C、D等3組配方品塗抹4週後,以B、C配方的的保濕效果最佳,其保濕率成長百分比分別為33.3%與31.7%。在皮膚彈性測試部份,B、C、D等3組配方成長百分比分別為5.4%與5.6%及4.3%。綜合實驗結果顯示,微脂粒經由超音波/冷凍乾燥後,其粒徑的大小與與使用乳化均質法並無絕對的相關聯。此外,銀耳所含之多醣體如製備成凍晶微脂粒後,能促進經皮吸收達到滲透的效果。

In this research, lyophilized liposomes were prepared from the extract of "Tremella fuciformis" with thin film/emulsification homogenization/ultrasound sonication followed by lyophilization. The experiments included measurement of the size of the liposomes in addition to evaluating the efficacies of three kinds of reconstituted lyophilized liposomes on skin hydration and elasticity. It was found that the particle sizes of formulas A and B produced by thin film/emulsification homogenization were 1563 ± 164nm and 1503 ± 415nm, respectively. On the other hand, the particle sizes of formulas A and B produced by thin film/emulsification homogenization/ultrasound sonication were 262.2 ± 4.90nm and 186.6 ± 9.53nm, respectively. Formulas B and C showed the best effects on skin hydration when the water content of horny layer was measured following application of formulas B, C, and D for four weeks; the hydration rates increased by 33.3% and 31.7%, respectively. Formulas B, C, and D showed an increase in skin elasticity by 5.4%, 5.6%, and 4.3%, respectively. The above findings showed that the size of liposome is not directly related to emulsification homogenization after ultrasound/lyophilization. Furthermore, polysaccharides in "Tremella fuciformis" can promote penetration through percutaneous absorption in the form of lyophilized liposome.
