p53-Dependent Expression of Ribosomal Protein S27-Like in Colorectal Cancer

背景:很多與p53相關的核醣蛋白已被證實與癌症有相關性,且當細胞遭遇到基因毒性時,RPS27L可以被p53誘發,但目前此誘發的RPS27L在大腸直腸癌中的角色所知甚少。方法:本研究使用組織免疫化學染色及西方點墨法,分析人類大腸直腸癌的組織中p53對RPS27L表現的影響,想了解RPS27L與p53的關係是否因p53DNA結合功能區序列改變而受影響。結果:在大腸直腸癌細胞株和組織中確認p53對RPS27L的表現關係。結論:本研究發現在晚期的大腸直腸癌病人中,RPS27L的表現與p53相關。其中有p53突變的大腸直腸癌病人中發現RPS27L的表現異常,而且會導致腫瘤細胞聚集,使得這些病人發生預後不良的情形。

Background and purpose: Many p53-associated ribosomal proteins (RPs) have been evaluated as cancer-related molecules. Although ribosomal protein S27-like (RPS27L) was shown to be a p53-inducible modulator that responds to genotoxic stress, little is known about its expression in colorectal cancer (CRC). Methods: We used immunohistochemistry and a Western blot assay to investigate alterations of RPS27L's expression by p53 in human CRC tissue specimens. The correlation between the expression of RPS27L and the p53 status was addressed by determining the sequence of the DNA-binding domain on the p53 gene. Results: The dependence of the expression of RPS27L on p53 was confirmed in colonic cell lines and tissues. Conclusions: Our findings show that patients with higher stages of CRC express RPS27L in a p53-dependentmanner. In CRC patients, mutant p53 seems to cause aberrant RPS27L expression, which may lead to the accumulation of tumorigenic cells without the regular activities of wild-type p53; these patients appear to have a poor prognosis.
