Indomethacin對懷孕中期胎鼠心肺之影響

摘要


目的:了解indomethacin對懷孕中期(12-18天)胎鼠的心肺血管之影響。方法:母鼠懷孕第12天開始,每天注射一次indomethacin(每公斤1毫克),至懷孕第18天,共七天。在胎鼠足齡時(母鼠懷孕第21天),對母鼠施行部腹產,取胎鼠,置於acetone與冰中,快速冷陳胎體至攝密-80º後,再以freezing microtome對胎體胸腔以每0.5-1毫米作橫向病理切片,再以顯微鏡對每一切割面的胎兒心肺血管解剖面作判讀,並計算其胸腺,心、肺容積,心室實質量之變化。結果:經Indomethacin處理後之實驗組胎鼠的體重,胸重腺容積與肺實質量分別減少為對照組的85%,62%,與82%,其心臟血管之大體解剖正常,但整體大血管之直徑明顯減小,動脈導管直徑僅為對照組胎鼠的77%,左心房容積減少為80%,且右心室實質量減少為85%,其具有統計上顯著之異,即使產前停藥3天,這些對胎兒產生的副作用也未恢復正常。結論:Indomethacin對懷孕中期胎鼠器官組織如胸腺,心肺血管的成長,有顯著影響。

Objective: to evaluate the influence of indomethacin on the cardiovascular system of fetal rats during the midtrimester of pregnancy. Methods: A daily dose of indomethacin, 1mg/kg injected into pregnant rats from the 12th day through the 18th day of pregnancy. Non-treated preqnant rats at the same gestational days served as controls. The fetal cardiovascular and pulmonary systems were studied on the 21st day of pregnancy (at term) using a rapid whole body freezing technique and then photographing of cross-sections of the frozen thorax, Results: There were no intrauterine fetal deaths in the treated pregnant rats. There were no signs of hydrops fetalis or congenital anomalies in the treatment group for mild decreased body weight to 85% of control group. The masses of thymus and lungs of the treatment group were notably decreased respectively to 62%, 82% of control group. Although no major cardiac structure anomalies were found contracted ductus arteriosus was noted in 15 fetal rats of the treatment group. Fetal cardiovascular morphometry in the treatment group revealed: decreased RV mass to 85% and decreased LA volume to 80% compared with controls. These data indicated that the side effects of indomethacin on off spring were persistent till birth even though the drug treatment was discontinued 4 days before delivery. Conclusion: Our data demonstrated that indomethacin has a significant effect on the growth of fetal organs, which included thymus, ung and cardiovascular system. (Full text in Chinese)
