Rank Regression Methods for Mapping Quantitative Trait Loci

摘要


大部分定位數量性狀基因座(QTL)的方法需要知道表現型的分配,例如常態、對數常態與韋伯。當對表現型分配有疑問而不能確定時,這些方法可能會不適用。本文以半母數加速失敗時間模式(semiparametric accelerated failure time model, Wei 1992)來描述QTL對會有設限失敗時間的表現型之影響,並利用秩有關的推論(rank-based inference)與經驗概似函數(empirical likelihood),發展出估計迴歸參數與偵測染色體上QTL位置的方法,並由一些模擬研究來評估所建議方法之表現。模擬結果顯示出QTL位置與影響效果的估計量是接近不偏的,而且當標識基因數增加時會更有效率。此外,當所建議的半母數模式分析法與Tzeng and Chiu (2006)所提出的有母數模式分析法比較時,本文的方法將具有強韌性(robustness)的效果。

Most of approaches for mapping quantitative trait loci (QTL) require a well-known phenotype distribution, e.g., normal, lognormal, or Weibull. These approaches may not be applicable when the phenotype distribution is doubtful and unspecified. In this paper, using the semiparametric accelerated failure time model (Wei 1992) for the effect of QTL on the censored failure-time phenotype, we developed approaches utilizing rank-based inference and empirical likelihood in estimating regression parameters and searching the entire chromosome for QTL. Some simulation studies were conducted to evaluate the performance of the proposed methods. Results from the simulated data indicated that both the proposed estimators of the QTL location and the QTL effects had only a little bias, and the efficiency would increase as the numbers of marks increased. In addition, the proposed semiparametric modeling strategy would obtain desirable robustness properties when compared with the parametric modeling strategy proposed by Tzeng and Chiu (2006).
