選擇性COX-2抑制劑Nimesulide作用於消化系統之藥理研究

Pharmacological Study of Selective COX-2 Inhibitor Nimesulide on Digestive System

摘要


Nimesulide is a non steroidal anti-inflammatory drug (NSAID) with selective inhibition of COX 2, and its consumption is increasing recently. Clinical use of selective COX 2 inhibitors superior to traditional NSAIDs are due to there less gastrointestinal side effects. The study purpose is to understanding the Nimesulide's properties: 1) analgesic effect .2) Effect of mall intestinal transportation capacity. 3) Effectively to gastric juices volume and the gastric juices contain free hydrochloric acid amount and the total acidity. 4) Effectively to stress gastric ulcers. The study objects are health male Wister rats about 8 weeks age. All investigational drugs are from Sigma Company and experimented by oral feeding. Experiment group: Nimesulide 100、200mg/kg. Contrast group: Salicylic acid 250mg/kg; Acetaminophen150 mg/kg; Mephenamic acid 200mg/kg. Control group: Dist H2O 5 ml/kg. The experiment results are as following: 1) analgesic effect: Salicylic acid 250mg/kg group after 180min has significant analgesic effect (P<0.01); Acetaminophen 150 mg/kg group after 120、180min has significant analgesic effect (P<0.01); Mephenamic acid 200mg/kg group after 60、120min has significant analgesic effect (P<0.01); Nimesulide 100/kg group after 360min has significant analgesic effect (P<0.01); Nimesulide 200/kg group after 60、120、180、240、360min has significant analgesic effect (P<0.01). 2) Effect of mall intestinal transportation capacity: all groups compared with Control group (Dist H2O) have little suppressed effect, but no significant difference 3) Effectively to gastric juices volume and the gastric juices contain free hydrochloric acid amount and the total acidity: about the total acidity, only Nimesulide group compared with Control group (Dist H2O) had no significant difference. About free hydrochloric acidity: Groups Nimesulide 100/kg and Nimesulide 200/kg have significant decreasing degree (P<0.05), the decreasing degree of free hydrochloric acidity is associated with gastric ulcers. 4) Effectively to stress gastric ulcers: Group Nimesulide 100mg/kg, shows the significant decreasing length of peptic ulcer than control group (Dist H2O), the length of peptic ulcer is 118.4±1.25mm (p<0.05). Conclusion: selective COX-2 inhibitors are not only good analgesic effect but also stress gastric ulcer suppressed effect.
