細胞培養系統生產絞股藍皂苷之研究

Production of Gypenosides by Cell Culture System of Gynostemma pentaphyllum (Thunb.) Makino.

摘要


絞股藍為葫蘆科絞股藍屬植物,絞股藍皂苷(gypenosides)則是其重要活性成份之一。本研究完成建立絞股藍皂苷測定流程,並於1種本地採集材料測得絞股藍皂苷成份。在組織培養方面,葉片培植體培養於含有0.2~1.0 mg/l picloram之培養基皆有癒傷組織生成,0.2 mg/l picloram與低濃度BA(0~1 mg/l)組合之培養基同時有不定根生成。癒傷組織繼代培養於含有5 mg/l BA及0.2 mg/l picloram之基礎培養基,一個月後其鮮重增加約2.57倍,癒傷組織呈現黃白色、緊實之型態。液體培養至第7~10天後細胞量不再增加,細胞內容物充實,且細胞呈現團聚現象。

Gynostemma pentaphyllum (Thunb.) Makino belongs to the Cucurbitaceae and its gypenosides closely resemble in ginsenoside. We established the GRg1 detection system and also found the similar component in a plant material native in Taiwan. Calli were induced from leaf explants on Murashige and Skoog (MS) basal medium plus 0.2-1.0 mg/l picloram. The first of these was pale yellow in color, compact in texture and watery in appearance. These calli could be maintained by subculturing every month with basal medium supplemented with 5 mg/l BA and 0.2 mg/l picloram. Liquid culture system was established by subculturing cells every 7-10 days.
