This study was to investigate the most suitable medium for the shoot tip culture of the Oncidium. Gower Ramsey 'Volcano Queen'. The explants with two leaf-primordia were harvested from terminal bud, the first lateral bud and the second lateral bud from the top. Comparing effects of the different salt levels and growth regulators in primary culture, shoot tips were cultured on solid medium with full strength Murashige and Skoog (MS) containing 1 mg/l NAA, 0.1 mg/l Kinetin, 20 g/l sucrose and 9 g/l agar possessed the best effect in survival rate of explants and numbers of PLB after 60 days. Furthermore, decapitated PLBs cut vertically into 4 segments were compared on the proliferation rate in different salt levels and coconut water percentages. The solid medium with full strength MS containing 20-25% coconut water have the highest proliferation rate.
