摘要

本研究主要探討文心蘭Onc. Gower Ramsey　‘Volcano Queen　’莖頂組織培養之最適當培養基。培植體取自頂芽、第一及第二側芽具二原體之生長點，比較不同鹽類濃度和生長調節物質對其初代培養之影響。培養60日後以含1 mg/l NAA、0.1 mg/l Kinetin、20 g/l蔗糖及9 g/l洋菜之全量鹽類濃度MS培養基在培植體存活率與類原球體數目上獲得最好的效果。此外，比較PLB去頂縱切成4片之培植體，在不同鹽類濃度和椰子水含量培養基中之增殖情形，結果以含20～25%椰子水之全量鹽類濃度MS固體培養基之增殖率最高。

This study was to investigate the most suitable medium for the shoot tip culture of the Oncidium. Gower Ramsey 'Volcano Queen'. The explants with two leaf-primordia were harvested from terminal bud, the first lateral bud and the second lateral bud from the top. Comparing effects of the different salt levels and growth regulators in primary culture, shoot tips were cultured on solid medium with full strength Murashige and Skoog (MS) containing 1 mg/l NAA, 0.1 mg/l Kinetin, 20 g/l sucrose and 9 g/l agar possessed the best effect in survival rate of explants and numbers of PLB after 60 days. Furthermore, decapitated PLBs cut vertically into 4 segments were compared on the proliferation rate in different salt levels and coconut water percentages. The solid medium with full strength MS containing 20-25% coconut water have the highest proliferation rate.
