醋酸菌Gluconacetobacter xylinus轉殖類血紅素蛋白基因(vhb)對細菌纖維素產量之影響

摘要


本研究的主要目的是希望藉由轉殖類血紅素蛋白基因(vhb),增加醋酸菌株在細菌纖維素的表現量。針對醋酸菌BCRC 12334、12335、14101、14148及14182等菌株,測試各菌株細菌纖維素的產量,以及對於抗生素的抗性等,發現菌株BCRC 12334具有較佳的纖維素產量與適合的抗生素敏感性,可供轉殖試驗之用。接著我們建構類血紅素基因表現載體並完成定序分析後,將該表現載體轉型入BCRC 12334醋酸菌株內,得到重組菌株G. xylinus VHb。利用西方墨點法進行蛋白質表現分析,可分別在分子量大小為16.0 kDa處發現預期的蛋白質條帶,進一步測試轉基因醋酸菌在細菌纖維素產量上的變化,結果轉型菌株G. xylinus VHb較野生型菌株BCRC 12334的纖維素產量增加21.7%。

This study is aimed to increase the cellulose production of Gluconacetobacter xylinus. G. xylinus strains BCRC12334, BCRC12335, BCRC 14101, BCRC 14148 and BCRC14182 were screened for their capability of cellulose production and antibiotic sensitivity. As a result, G. xylinus BCRC12334 was chosen for gene manipulation. Plasmid containing vhb under the control of the tac promoter was constructed and transformed into G. xylinus BCRC 12334. The recombinant strain was able to produce the encoding protein with the expected molecular weight of 16.0 kDa as analyzed by Western blotting. Consequently, we have found that the cellulose production of recombinant G. xylinus VHb was about 21.7% higher than that of wild type.
