大豆發酵液乾燥粉末萃取物具有抑制血管新生的功效

Extracts of Dried Powder of Liquid Derived from Soybean Fermentation Inhibit Angiogenesis

摘要


血管新生(angiogenesis)是指在生理上由原來已存在的血管分化新的血管而發展成一個血流供應系統的過程,而腫瘤細胞生長過程中,能夠產生誘導血管新生的生成因子〔如血管內皮生長因子(vascular endothelial growth factors,VEGFs)〕,促使血管內皮細胞進行血管增生作用,新生的血管可為腫瘤的生長提供營養和氧氣,促進腫瘤細胞快速生長及侵襲其他組織。因此,抑制腫瘤細胞的血管增生可作為配合化學療法之抗癌的重要策略,相關研究已受到全世界科學家的重視。在本研究中,首先以大豆發酵液乾燥粉末之水萃取物及乙醇萃取物利用大鼠動脈環分析模式探討動脈血管增生的影響。結果發現,水與乙醇萃取物皆可顯著抑制大鼠主動脈環血管增生,並具有劑量效應(dose-dependent)相關性,其中乙醇萃取物抑制血管增生的效果優於水萃取物。研究結果亦指出大豆發酵液乾燥粉末及乙醇萃取物可抑制人類肺癌細胞A549異體移植裸小鼠之腫瘤的生長及血管增生作用,其作用機制推測可能與抑制VEGF、VEGFR-2及PCNA蛋白質的表現密切相關。

Angiogenesis is the process of vascular growth by sprouting from preexisting vessels. It is a complex process involving secretion of angiogenic factors by tumor and host cells; endothelial cell proliferation, migration, and invasion; extracellular proteolysis; capillary formation; and vessel remodeling. Recently, inhibition of tumor angiogenesis has become a clinical anti-cancer strategy in conjunction with chemotherapy. In the present study, extracts of dried powder of the liquid derived from soybean fermentation on angiogenesis was investigated. Results revealed that both water and ethanol extracts of such significantly inhibited new vessel formation in the rat aortic ring assay in a concentration dependent manner; ethanol extract was found to be more effective than water extract. Both dried powder and ethanol extracts were also found to inhibit tumor growth and angiogenesis in the A549 lung cancer xenograft model, probably by inhibiting the production of VEGF, VEGFR-2, and PCNA.
