台灣本島硬葉馬尾藻褐藻醣膠粗萃物於抗癌之研究

Anti-cancer Research of the Crude Fucoidan Extracts of Sargassum aquifolium from Taiwan

摘要


從褐藻萃取的褐藻醣膠已被文獻證實具有抗癌能力,因此褐藻醣膠相關的研究逐漸增加。本研究的主要目的是使用從台灣屏東五個地區採集到的硬葉馬尾藻進行酸萃取,萃取出來的褐藻醣膠進行一小時和兩小時萃取後的性質和生物活性。本次實驗證實了相同海域但生長在不同位置的硬葉馬尾藻的產率和硫酸根含量有明顯差異,但是其總醣量無明顯的不同。經過HPLC的分析後,本次萃取出的褐藻醣膠的單醣組成為甘露醣、葡萄醣、半乳醣、木糖和岩藻醣。在進行細胞存活率測試時,發現船帆石(B04)區採集到的硬葉馬尾藻萃取出的褐藻醣膠在進行兩小時的酸水解後,當濃度為200 μg/mL下,癌細胞HCT116的存活率為58.12%。本次研究也發現在台灣-屏東-船帆石採集到硬葉馬尾藻,其萃取出的褐藻醣膠具有良好的抗癌能力。

Fucoidan extracted from brown algae has been proven to have anti-cancer ability in previous studies, so the research fields on fucoidan are growing wildly in recent years. In this study, we collected the Sargassum aquifolium from five regions in Pingtung, Taiwan, and used the acid extraction method to extract fucoidan, then investigate the properties of the biological activities of the fucoidan after hydrolysis for an hour and two past the extraction. This experiment confirmed that the extraction yield and sulfate content of Sargassum aquifolium grown from different locations in the same sea area were significantly different, and the fucoidan total sugar content has no significant difference. The monosaccharides compositions are mannose, glucose, galactose, xylose, and fucose. During the cell viability test, the fucoidan extracted from Sargassum aquifolium, collected in Chuanfan Rock (B04) area was acid hydrolyzed for two hours before the test. The result shows that, at a concentration of 200 μg/mL, the cancer cell HCT116 survival rate was 58.12%, which exhibits decent cytotoxicity to cancer cell. This study found that the fucoidan extracted from Sargassum aquifolium collected from Chuanfan Rock, Pingtung, Taiwan, has significant anti-cancer ability.
