本研究主要為評估化妝品常用之二十六種精油對於清除ABTS^(.+)自由基(2,2'-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid))、還原能力(Reducing power)之抗氧化活性及其不同反應時間時之抗氧化活性差異,並探討主要之抗氧化活性成分。結果顯示二十六種精油中以黑胡椒(Piper nigrum)精油之抗氧化活性為最佳,當其精油濃度為1.0mg/mL時,能清除90.88%的ABTS^(.+)自由基,其半數有效抑制濃度(EC_(50))為101.43μg/mL,於反應時間方面,反應10分鐘時之ABTS^(.+)自由基清除能力,為反應60分鐘時的97.82%;於還原能力方面,黑胡椒精油濃度為5.0mg/mL時,其還原能力為標準品BHA之114.79%,並於反應時間方面,反應20分鐘時之還原力,為反應60分鐘時的99.26%。最後,以氣相層析質譜儀(Gas Chromatograph-Mass Spectrometer, GC-MS)鑑定其主要之化學組成分,發現黑胡椒精油中含有高比例的丁香酚(Eugenol),含量為59.02%。綜合以上之結果,證明黑胡椒精油中的丁香酚為主要清除ABTS^(.+)自由基及提供還原能力之主成分,且其於反應10分鐘時,抗氧化活性較其他精油為佳。再者,抗氧化活性愈佳之精油,其活性受到反應時間之影響則越小。
This study is major in the different reaction time of the commonly used twenty-six kinds of essential oils for the scavenging ABTS^(.+) free radical and reducing power activities in cosmetics and exploring their main chemical constituents. The results were shown that the black pepper oil shows the most significant antioxidant capacity in twenty-six kinds of essential oils in 10 minutes during the reaction time and the better the antioxidant capacity of the oil; the less reaction time is affected. In the concentration of 1.0 mg/mL of black pepper essential oil, 90.88 % of ABTS^(.+) radical was removed and the EC_(50) is 101.43 μg/mL. 97.82 % of ABTS^(.+) free radical scavenging ability has been shown in 10 minutes during the 60 minutes reaction time. In the concentration of 5.0 mg/mL of black pepper essential oil, it was shown the 114.79 % reducing power relative to the standards of BHA. 99.26 % of reducing power has been shown in 20 minutes during the 60 minutes reaction time. The main chemical components of the black pepper essential oil were identified by gas chromatography mass spectrometry (GC-MS) and a high proportion of 59.02 % of eugenol in it.
