圈養領角鴞血液生化、蛋白質電泳參考值之建立及研究

The Study in Haematologic, Plasma Biochemical and Protein Electrophoretic Values of Captive Collard Scops Owls (Otus bakkamoena)

摘要


血液生化參考值在臨床醫學上提供臨床獸醫師動物個體在營養、生理狀況的判讀資訊以及臨床疾病的診斷依據,是野生動物救傷及復育工作中極為重要的基礎研究,然而目前台灣本土物種的相關資訊相當有限。 領角鴞(Otus bakkamoena)屬於保育等級第二級珍貴稀有保育類野生動物,在台灣是與人類生活環境最接近的鴟鴞科鳥類,然而隨著大量棲地的破壞,野生族群數量受到影響而漸減,領角鴞每年在台灣救傷單位占野鳥救傷數量的比例很高。本研究目的為建立領角鴞的血液生化參考值及蛋白質電泳參考值,並進一步評估年齡、性別及體態在各項數值中的差異性,以期在未來可以應用於本土野鳥救傷復育的臨床工作。 本研究共收集圈養領角鴞亞成鳥25隻、公鳥21隻及母鳥15隻,以物理保定進行肱靜脈採血,血液常規檢驗項目包括血容比(PCV)、血紅素(Hb)、紅白血球計數、血球分類比例,及血液生化學項目包括總蛋白(TP)、白蛋白(Albumin)、葡萄糖(Glucose)、麩胺醯轉移脢(GGT)、乳酸脫氫酶(LDH)、天門冬酸鹽轉胺酶(AST)、膽固醇(Cholesterol)、肌酐激脢(CK)、尿酸(Uric acid)、血鈣(Ca)、血磷(P)、血鈉(Na)、血氯(Cl)、血鉀(K)等項目,建立健康個體參考值的範圍,另外以血漿進行蛋白質電泳分析,呈現各蛋白質種類的比例及數值參考值。 結果顯示年齡及性別在領角鴞多項血檢數值造成影響,成鳥的血容比、血紅素、異嗜球與淋巴球比例(H/L ratio)、總蛋白、白蛋白、白蛋白與球蛋白比例(A/G ratio)、膽固醇、血鈉及血氯高於亞成鳥;亞成鳥的血磷及白血球數量顯著高於成鳥;麩胺醯轉移脢於不同年齡有顯著差異;而母鳥的天門冬酸鹽轉胺酶高於公鳥有差異趨勢;體態指標(body condition index)則與各項血檢項目無顯著相關,顯示年齡與性別的確會造成領角鴞部分血檢項目的影響。在領角鴞的各種血球形態上與其他鴟鴞科鳥類相似,白血球分類以淋巴球為主,異嗜球次之。 本研究的結果建立領角鴞的血液生化學及蛋白質電泳的參考數值,對於領角鴞的臨床病理學、醫療救傷及保育復育工作有重要的價值,本實驗模式也可應用於其他本土鳥種之相關研究。

Lots of valuable medical information of avian species can be obtained from hematology and blood biochemistry such as nutritional status, physical condition and also the diagnosis of diseases. Studies of blood chemistry and hematology are the most important database for wildlife rescue and conservation. However, this kind of studies for endemic species in Taiwan is rare. Collared Scops Owl(Otus bakkamoena)is one avian species living closely to people and listed on the Wildlife Conservation Law as a rare and valuable species in Taiwan (Wildlife conservation law, 2009). The wild population of this owl keeps declining due to nature habitat destruction. Collared Scops Owls are the major species of raptor rescued cases in wildlife rescue centers around Taiwan. The aim of this study is to establish the hematologic and biochemical references with protein electrophoretic fractions in clinically healthy Collared Scops Owls. Blood samples were taken from the brachial vein from 25 subadults, 21 adult males, and 15 adult females. Hematologic values included packed cell volume(PCV), hemoglobin(Hb), red blood cell(RBC)count, white blood cell(WBC) count, and leukocyte differential count. Biochemical analyses included total plasma protein, albumin, glucose, Gamma glutamyltransferase(GGT), Lactate dehydrogenase (LDH), Aspartate aminotransferase(AST), cholesterol, Creatine kinase (CK), uric acid, calcium, phosphorus, Sodium, Chloride and potassium concentrations together with protein electrophoretic fractions. Adults had higher levels of PCV, Hb, H/L ratio, total protein, albumin, cholesterol, A/G ratio, Na and Cl than subadults. Subadults had higher levels of phosphate concentration than adults. Subadults and adults had different levels of GGT. Females had higher levels of AST than males but not significantly. However, body condition index had no effects on these analyses. Morphology of blood cells from the Collared Scops Owls are similar to other Strigidae owls. Lymphocytes and heterophils are the most abundant cellular type. The results of the hematological and biochemical reference values and protein electrophoresis presented in this study can be directly used in the clinical veterinary medicine and wildlife conservation in this species. This established research model of avian clinical pathology also contributed to the future work of other endemic avian species in Taiwan.
