摘要


端粒在多數癌症細胞當中因具有過度表現的端粒酶而受到延長,因此成為染色體上的重要腫瘤標的。文獻中設計配體穩定端粒形成四股結構,造成端粒酶功能受抑制而限制腫瘤成長。其中配體BMVC具有辨識腫瘤並穩定端粒鳥嘌呤四股結構的作用及其螢光特性,本論文在結構上積極地引入具有光激活的端粒傷害分子片段,使在顯影出癌細胞的位置同時也伴隨有毒殺癌細胞的潛力。在文獻報導中得知, BMVC一側的吡啶陽離子與四股結構平行股上的胸腺嘧啶T17距離3.75 Å,並發生

Telomeres, an important tumor chromosomal target, are enlogated by overexpressed telomerase in most tumor cell. Researchers have attempted to design and synthesize G-4 ligands to stabilize telomeric G-quadruplex aiming for telomerase inhibition as a potential anti-tumor route. Taking advantages of the strong binding of BMVC with telomeres as well as its fluorescent sensing, we decided toreplace one of the pyridinium ring with a photoactive thymine in order to damage the telomere G-quardruplex.
