評估啤酒花萃取物之抗菌性與及抗發炎性

Evaluation on anti-microbial and anti-inflammatory effects of hops extract

摘要


啤酒花於啤酒釀造中賦予啤酒苦味和風味,近年發現異戊二烯基的黃烷酮類成份,具有許多生物活性。而痤瘡桿菌 (Propionibacterium acnes) 與皮屑芽孢菌 (Malassezia furfur) 對尋常性痤瘡 (Acne vulgaris) 及頭皮屑 (dandruff) 的生成扮演重要的角色。本研究利用有機溶劑萃取啤酒花,將其區分為水相分萃物 (Aqueous fraction, W) 及乙酸乙酯相 (Ethyl acetatefraction, EA) 分萃物,探討其對兩種微生物的抑制性,再以Lipopolysaccharides (LPS) 1 μg/mL、P. acnes 500 μg/mL 和M. furfur 500 μg/mL分別誘導THP-1細胞產生發炎反應,並測定其誘導生成之NO產量及對生成細胞激素TNFα及IL-8的影響。抗微生物作用的結果顯示,抑制P. acnes生長的最小抑制濃度 (MIC) 和最小殺菌濃度 (MBC) 為8 μg/ml和64 μg/ml的乙醇水相分萃物(EOW);而可抑制M. furfur生長的MIC為所有EA分萃相在1000 μg/ml皆可抑制。更進一步探討其對發炎作用的影響,乃分別以LPS和微生物誘導THP-1細胞發炎使其產生NO,EOW和乙醇的乙酸乙酯相分萃物(EOEA)為16 μg/ml其NO產量抑制率分別與正控制組(+)相比皆達顯著差異(P < 0.001)。此外,以LPS和微生物誘導THP-1細胞生成促發炎細胞激素TNFα發現EOW可顯著使TNFα抑制率皆顯著高於正控制組 (P < 0.001),但EOEA僅對P. acnes誘導者具有抑制效果。以LPS和微生物誘導生成IL-8結果顯示EOW及EOEA濃度為16 μg/mL即可顯著降低對LPS誘導和P. acnes組之IL-8生成量(P < 0.05)。由本實驗結果可發現啤酒花乙醇萃取的水相分萃物(EOW)濃度約在16 μg/mL可以兼具抑制痤瘡桿菌生長及減輕發炎現象的效果,顯示啤酒花萃取物具未來開發為相關生技產品之潛力。

Hops, the female inflorescences of the hop plant (Humulus lupulus L.), are widely used in the brewing industry to add bitterness and aroma to beer. Recently, some flavanones with prenyl groups have been identified in hops and beers, and it had many biological activities. Propionibacterium acnes and Malassezia furfur play important roles in this two common skin diseases of Acne vulgaris and dandruff. The object of this study was to investigate different extracts of hop cone that effected anti-microbial and anti-inflammatory activities. In this study, LPS ( 1μg/ml)、P. acnes ( 500 μg/ml)or M. furfur (500 μg/ml) were co-treated with THP-1 cell to evaluate the production of pro-inflammatory factors. The result revealed that the lowest MIC against P. acnes was 6 μg/ml extracted by aqueous fraction of 95% aqueous ethanol (EOW) and MBC was 64 μg/ml EOW, whereas the lowest MIC against M. furfur was 1000 μg/ml extracted by ethyl acetate fraction of several solvents. In addition, 16 μg/ml of EOW and EOEA were presented significantly inhibitory abilities to nitrite oxide (NO) production (P < 0.001). All inflammatory induced group decreased the TNFα production by co-treating with 16 μg/ml EOW, but the TNFα production reduced by 8 μg/ml EOEA only in P. acnes induced group. Moreover, the concentrations to decrease IL-8 production are 16 μg/ml EOW and EOEA at LPS and P. acnes induced group, but all increased at M. furfur induced group. In conclusion, aqueous fraction of 95% aqueous ethanol (EOW) had better bioactivity for anti-microbial effect against P. acnes and alleviated the inflammatory effect.
