五味子及芝麻被發現對於四氯化碳(CCl4)所誘發肝損傷之老鼠具有保護的作用。本研究擬探討五味子與芝麻混合物之抗氧化力及抗發炎的效果,另外以人體試驗探討護肝之功能,並測定對人體抗氧化能力與抗氧化酵素活性之影響。 人體試驗共計40位肝指數偏高(GOT或GPT介於40~100U/L)之健康受試者全程參與,採隨機、雙盲、安慰劑組對照進行。本試驗將受試者分為兩組,一組服用五味子與芝麻之混合物,另一組則服用安慰劑,試驗前的一個月為run in period,試驗期為五個月,試驗期後的一個月為follow up period。受試者每天服用兩次的試驗樣品或安慰劑,每次服用兩錠,一次為午餐飯前,一次則為晚餐飯後。試驗前後進行體位測量,每個月採集血液檢體,並於試驗之起始,及第三、五個月進行腹部超音波檢測。 結果顯示,連續服用五味子及芝麻混合物五個月後,可以顯著降低血漿glutamate oxaloacetate transaminase(GOT)及glutamate pyruvate transaminase(GPT),延長血漿中低密度脂蛋白(low density lipoprotein;LDL)氧化延遲時間,提昇受試者體內總抗氧化能力和降低脂質過氧化反應,並顯著提高紅血球中glutathione peroxidase(GSH Px)及glutathione reductase(GSH Rd)之抗氧化酵素活性。
Schisandra and sesame were found protective from carbon tetrachloride (CCl4) induced liver damage on rats. This study was focused on the anti-oxidant capacity and anti-inflammatory effect of the mixture of schisandra and sesame. A human clinical trial was used for its protection from liver inflammation, and also determined the antioxidant capacity and enzyme activities. Forty healthy subjects were completed involved in this study, whose liver function index were slight high (GOT or GPT in the range of 40 to 100 U/L). This was a randomized, placebo controlled and double blind design. We randomized subjects into two groups. One group of subjects took the mixture of schisandra and sesame tablet, the another group took placebo. One month before administration was run in period. Experimental period was five months, one month after the stop of administration was follow-up period. Each subject took the mixture of schisandra and sesame tablet or placebo twice a day. Two tablets were taken before lunch, and the other two were after dinner. Anthropometric measurements at initial and end of study were determined. Blood collection every month, and abdominal ultrasounic examination were performed at the initial, 3rd and 5th month. Five months of continuous administration of the mixture of schisandra and sesame tablet could significantly lower GOT and GPT and prolonging the lag time of LDL oxidation. In addition the total antioxidatant activity, GSH Px and GSH Rd were greatly increased.
