花生膜萃取物之調節體脂肪與抗氧化效果

Effect of body fat regulation and antioxidant activity from peanut skin extract

摘要


WHO&FDA於1996年已經將肥胖列為慢性疾病之一。花生膜萃取物中含有豐富的酚類物質,能增強人體內抗氧化能力,並減緩人體內脂肪之形成。本研究分為兩部分,目的為確認花生膜萃取物之功效性成份與減少體脂肪功能和抗氧化能力之探討。第一部分為體外實驗,測定受試物之總酚與總類黃酮含量以及使用HPLC測定其有效成分。第二部分為人體試驗,此試驗共募集了36位BMI≧27或體脂肪大於 30% 的受試者,隨機分成兩組,每天午、晚各食用2顆受試物或與樣品相仿之安慰劑,持續6週,最後完成實驗者為28位。試驗期間每3週檢測體重、體脂肪變化,並採集血液測定血糖、血脂變化、GOT、GPT等生化指標以及測定血液中抗氧化能力,並於使用產品前後隨機抽選5位受試者配合核磁共振掃描檢查腹部脂肪層分布及厚度,觀察補充花生膜萃取物是否具有減少體脂肪的效果。研究結果發現,以酒精萃取受試物後,發現總酚含量為32.01 ± 0.01 mg GAE / g ,類黃酮為 77.41 ± 0.01 mg CE / g ,而DPPH清除率為72.12 %;總抗氧化能力為 0.69 ± 0.00 mM,進一步使用HPLC分析,發現受試物中具有兒茶素、阿魏酸、咖啡酸等酚類化合物。在人體實驗結果發現,連續食用6週受試物,實驗組與安慰劑組比較,體脂肪有顯著下降 5.2 %,且在血中低密度脂蛋白膽固醇也有顯著下降 6.25 %。在抗氧化方面,血漿中丙二醛有顯著下降30.5 % (p < 0.05)。另外血漿中總酚顯著上升 21.2 %,而血球中超過氧岐化酶顯著上升28 %,並且穀胱甘肽過氧化酶、穀胱甘肽還原酶皆有顯著上升分別為19.7 % 與16.5 % (p < 0.05)。綜合以上,食用花生膜萃取物具有減少體脂肪與提升體內抗氧化之能力,可能是因為花生膜萃取物中具有對身體有益的酚類物質。

WHO and FDA claim obesity is one of the chronic disease since 1996. Peanut skin extracts have abundant phenolics to enhance antioxidant activity of human body and prevent body fat formation. The study divide to two parts, one is analysis the component of sample by ethanol, the extract had effects on total extractable and capacity of antioxidant activity. The other one is evaluation effect of peanut skin extract consumption on regulation of body fat and antioxidant activity of the thirty-six volunteers who are BMI ≧ 27 or body fat over than 30% enrolled in this double-blinded random clinical trial. Subjects were randomly assigned to two groups. They were asked to consume peanut skin extract ( about 140.8 mg GAE / day ) with their habitual diet for 6 weeks. The subjects measure anthropometric values, body fats and collect blood parameter including blood sugar, lipid profiles, GOT, GPT and also measure antioxidant activity of the plasma and erythrocyte. The study results reveal total phenolics of ethanol extracts of sample was 32.01 ± 0.01 mg GAE / g. Total flavonoids was 77.41 ± 0.01 mg CE / g. About totally antioxidant activity and DPPH scavenging ability of ethanol extracts were 72.12 ± 1.30 mM and 72.12 %, respectively. Finally used HPLC identify functional compound found catechin, caffeic acid, and ferulic acid in sample. The second part that clinical trial showed body fat was significantly decrease 5.2% (p < 0.05) and LDL-C were significantly decrease 6.25% (p < 0.05 ) compare form initial period. In the antioxidant capacity the MDA was significant decrease 30.5% (p < 0.05 ) from plasma. In the other hand, SOD, Glutathione peroxidase and Glutathione reductase activity were significantly increase 28%, 19.7% and 16.5% (p < 0.05), respectively. In conclusions, intake peanut skin extract can decrease body fat and increase antioxidant capacity that functional effect contribute by phenolics.
