翼狀贅肉培養之纖維母細胞經脂多醣刺激後之長鏈非轉譯性核醣核酸變化

摘要


翼狀贅肉是外眼部常見且會進展的疾病,與日照中的紫外線有強烈相關。嚴重期別會導致視力受損、眼球運動受限、慢性發炎以及美觀問題。特性包括增生、不定形、具發展期別,有復發特質。其培養出之纖維母細胞的活性依據期別不一。非轉錄性核糖核酸(Noncoding RNA)之長鏈非轉錄性核醣核酸(Long noncoding RNA);簡稱Lnc RNA,具有基因調控的功用。本研究期能找出翼狀贅肉纖維母細胞裡異常表現的Lnc RNA種類,發掘出相關調控功能,與mRNA或其他轉錄出蛋白質之相關性。本研究收集自民國一零四年七月至民國一零五年七月於彰化秀傳紀念醫院接受翼狀贅肉切除手術之患者檢體。將取下之翼狀贅肉部分培養長出之纖維母細胞,使用脂多醣Lipopolysaccharides(LPS)刺激後,與未經LPS刺激比較核醣核酸序列(RNA sequencing)表現,運用基因本體論(Gene Ontology)方法,使用基因資料庫找出表現量改變之lncRNA。經LPS刺激前後,表現差異之LncRNA量,上升的有十五種,下降的有四種。而上升之mRNA有一百五十二種,下降有五十八種。經核糖核酸定序發現,與翼狀贅肉相關之mRNA多數與發炎途徑相關。而LncRNA之改變,實際的功能尚待探討。

Pterygium is a common ocular surface disease which related to ultraviolet exposure. The several characteristics of pterygium includes proliferation ,malformation ,recurrence and having progression stages. Furthermore, the activity of fibroblast cultured from pterygium varied according to stages. Long noncoding RNA is believed to having ability to regulate gene expression. Our study aims to define abnormal expressed long noncoding RNA in pterygium fibroblast, and discover its associated regulated mechanism. Our study recruit excised pterygium specimens from ChangHua Show Chwan Memorial hospital during July, 2015 till July, 2016. Afterward, fibroblast were cultured from those excised pterygium particle. We monitor the differential expressions between lipopolysaccharides-treated fibroblast and clear fibroblast at the RNA sequencing. Gene Ontology is applied for annotation for possible functions component, and involved biological pathway of long noncoding RNA and mRNA. 50 pterygium specimens were collected during the research period. After treating by lipopolysaccharides, we found there are 15 up-expressed, and 4 down-expressed long noncoding RNA; 152 up-expressed and 58 down-expressed mRNA.We found most of mRNA differential responses to lipopolysaccharide-treated mRNA are related to inflammation. Meanwhile, the variation of long noncoding RNA is to be determined.
