覆盆子萃取物抑制人類口腔癌細胞轉移和侵襲之機制探討

摘要


根據行政院衛福部的最新調查顯示在101年主要死因為癌症,且在過去十年內口腔癌的死亡率增加了15.8%,而造成口腔癌死亡率增加的主因為發生癌細胞轉移現象,因此抑制癌細胞轉移是現今重要的議題。癌細胞會藉著基質金屬蛋白酶 (matrix metalloproteinases-2,MMP-2) 的分泌來降解細胞外基質,而使癌細胞發生轉移現象,且已知透過MAPK pathway (mitogen activated protein kinase) 可以調控MMP-2的表現。覆盆子萃取物 (Rubus idaeus extract,RIE) 來自屬於薔葳科木本植物的覆盆子,其果實富含許多抗氧化物,有抗發炎、抗菌和降血壓等效用。在過去的文獻中指出覆盆子可以有效預防腎結石的形成以及有效的控制胃炎,甚至有文獻證實覆盆子在肺癌細胞中可以有效的抑制癌細胞的爬行能力因而抑制癌細胞的轉移,但覆盆子在口腔癌細胞中的抑制效果如何及訊息傳遞路徑尚未有文獻證實。本研究選用口腔癌細胞株 (SCC-9) 來進行實驗,發現在不同的覆盆子萃取物濃度 (0、25、50、75、100

According to the latest survey from the Ministry of Health and Welfare, the main cause of death in 2012 in Taiwan was cancer, and in the past decade, oral cancer mortality increased by 15.8%, the cause of which was primarily the occurrence of cancer metastasis. Therefore, the inhibition of cancer metastasis is currently a critical topic. Cancer cells secrete matrix metalloproteinases (i.e., matrix metalloproteinases-2, MMP-2), degrading extracellular matrix, and promoting cancer cell metastasis via mitogen-activated protein kinase (MAPK) pathway, which regulates the expression of MMP-2. Raspberry extract (Rubus idaeus extract, RIE) from woody plants belonging to the Rosaceae family is rich in antioxidants, has antiinflammatory and antibacterial properties, lowers blood pressure, and exerts other beneficial effects. According to the literature, pointed raspberries effectively prevent the formation of kidney stones and effectively control gastritis. In addition, raspberries have been confirmed to inhibit the crawling ability of lung cancer cells and thereby inhibit cancer metastasis; however, the inhibitory effect of rasperries in oral cancer cells and signaling pathways have yet to be confirmed.
