極低頻磁場對肝癌細胞株生長與移動力的影響

中文摘要 電磁場對環境的影響與時俱增。隨著生活、工作上的需求,常用頻段的電磁場逐漸增多、複雜,尤其極低頻磁場更是與我們接觸頻繁,籠罩在此環境下的人類細胞是否也因此逐漸變化中?過去的文獻中,正、反影響報導都有,然偏向部分射頻與正常人細胞較多。本研究以荷姆赫茲線圈產生極低頻均勻磁場,作用在肝癌細胞株HepG2、 Hep3B上,以螢光顯微鏡並多天觀察其生長及型態變化,發現在60Hz,8~10高斯作用區域有自噬(autophagy) 跡象,然後以HepG2細胞配合MTT assay、AO stain及加入autophagy之抑制劑3MA實驗,證實極低頻磁場能引發肝癌細胞autophagy,而DAPI stain 亦呈現HepG2細胞在極低頻磁場下其生長被抑制,apoptosis 是有參與的。又因HepG2細胞 wound healing實驗的不明顯,故以 Hep3B進行MMP實驗,結果MMPs的分泌表現可用以說明Hep3B wound healing的現象,也證實極低頻磁場會促進Hep3B 細胞的移動。本實驗中其標的細胞選以國內癌症死亡率居高位的肝癌細胞株進行實驗,期望能對肝癌之治療提供時機與有益的資訊。

英文摘要 The effect of electromagnetic field on our living conditions is increasing. The more you want to improve the quality of life and work, the more radio frequency bandwidths you need to have. In particular, the signals of extremely low frequency (ELF) bands surround us today. Are human cells being changed gradually in this situation? There were many contradicting reports in the past, but most focused on the relationship between a partial radio frequency (RF) range and normal human bodies. We used Helmholtz coil to produce an uniform magnetic field to treat the human hepatocellular carcinoma, HepG2 & Hep3B cell lines, then monitored the status of cell growth and morphological change with inverted fluorescent microscope over multiple days, and found signs of cell autophagy at the condition of 60Hz, 8~10gauss and 2 hrs exposure. It showed autophagy after HepG2 experiments of MTT assay, AO stain and autophagy inhibition assay by 3MA. With DAPI stain, it also provided a demonstration of apoptosis when HepG2 cells growth was inhibited after exposure to an ELF-10gauss magnetic field. Since HepG2 cells did not perform wound healing, we arranged MMP assay on Hep3B cells to explain how it promotes wound healing and found that in a condition of ELF magnetic field, Hep3B cells mobilized. These experiments were implemented on hepatocellular carcinoma cell line which is the top mortality of cancer in Taiwan. Hopefully it will provide information useful for today’s cancer therapy.
