本研究開發一套方法利用固相萃取法(Solid Phase Extraction)將尿液中會對螢光檢測產生干擾之螢光物質去除後,加入氧化態菸鹼醯胺腺嘌呤二核苷酸(NAD+)以及D-乳酸去氫酶將尿液中的D-乳酸反應為NADH並以波長為340 nm之紫外發光二極體激發NADH使其產生490 nm波長的螢光,並偵測其螢光訊號以量測出尿液中D-乳酸濃度之生物感測器。此檢測方法擁有良好的線性程度(R2=0.9979),其偵測範圍可從1 μM到125 μM。檢測準確度為: 93.22~110.54%及精密度為: 2.04~20.83%。其線性程度、準確度與精密度符合分析所需之規範。而且此檢測方法相較於二維高效能液相層析系統可以將單一檢品的檢測時間從90分鐘降低為1分鐘以內。
The purpose of this study is to develop a method using solid phase extraction to remove the impurities in urine to decrease fluorescent noise, then add the nicotinamide adenine dinucleotide (NAD+) and D-lactate dehydrogenase (D-LDH) in urine. D-LDH will catalyze D-lactate and NAD+ to pyruvate and NADH, and then using biosensor that use the wavelength of 340 nm UV-LED to excite NADH and release the fluorescence of 490 nm to detect the concentration of D-lactate in urine. This method has good linearity (R2=0.9979) and calibration range from 1 μM to 125 μM. Accuracy is between 93.22~110.54% and precision is between 2.04 - 20.83%. The linearity, accuracy and precision of this method are complied with rules of analysis. And compared to Two-dimensional HPLC system (2-D HPLC system) this method can reduce the determination time from 90 minutes to 1 minute each sample.
