中文摘要 論文名稱:肝癌細胞Lysyl oxidas 表現與致癌作用機制之探討 研究所名稱:台北醫學大學醫事技術系暨研究所 研究生姓名:莊千慧 畢業時間:九十六學年度第二學期 指導老師:何元順博士 (台北醫學大學醫學院醫學檢驗生物技術學系暨研究所教授) 肝癌是全世界排名第五名的惡性腫瘤,而酒精性肝炎與B 型肝炎 病毒是造成肝癌的主要原因。B 型肝炎病毒經由垂直感染造成新生兒為B 型肝炎病毒帶原者,酒精性肝炎主要是因酒精取得容易,長期酗酒所造成,二者皆會造成肝硬化,最後演變成肝癌。Lysyl oxidase 是一種氨基氧化酶,位在細胞外基質,當組織產生慢性發炎反應之後,組織會開始修復,這時候lysyl oxidase 產生去氧化膠原蛋白與彈性纖維,形成cross-linking ,使組織纖維化,漸漸形成肝硬化,慢慢演變成肝癌。因此我們希望能找出lysyl oxidase 與肝癌的相關性,經由本次研究結果發現,在臨床的肝癌組織切片中,免疫化學組織染色法發現lysyl oxidase 在肝癌細胞表現量比正常肝臟細胞表現量較多,接下來我們利用Hep 3B cell 每天加入10mM Ethanol 培養,lysyl oxidase 隨著長期酒精培養的代數的增加表現量就越高,接著合併使用天然的抗氧化物蓝莓的萃取物與薑黃萃取物,合併使用這二個抗氧化物會抑制lysyl oxidase 活化型蛋白。本次研究結果建議,lysyl oxidase 的表現的確與惡性肝腫瘤相關,經由體外實驗發現lysyl oxidase 會因長期使用酒精而增加,目前也找到天然的抗氧化物藍莓萃取物與咖哩萃取物可以抑制肝癌細胞分泌活化型的lysyl oxidase,期待應用在臨床上,減少肝硬化的發生。

Abstract Title of Thesis: Studies on the expression of lysyl oxidase in hepatocellular carcinoma cells and carcinogenesis Institule : Graduate Institute of Biomedical Technology, Taipei Medical University Author: Chuang-Chien Hui Thesis advised by: Yuan-soon Ho,Ph.D. (Professor, School of Medical Laboratory Science and Biotechmology,College of Medicine, Taipei Medical University) Hepatocellular carcinoma (HCC) is one of the most common cancers in the world with an annual incidence of more than 500000 in the year 2000.Alcohol excess is associated with a spectrum of disease ranging from simple steatosis through steatohepatitis to cirrhosis and, in some, hepatocellular carcinoma. Hepatitis B virus (HBV) infection is still the most common cause of hepatocellular carcinoma and liver cirrhosis world wide. Lysyl oxidase could regulated formation and repair of the extracellular matrix (ECM) by oxidizing lysine residues in elastin and collagen, thereby initiating the formation of covalent crosslinkages which stabilize these fibrous proteins. We found and elevated expression of the lysyl oxidase (LOX) in biopsies of Hepatocellular carcinoma (HCC) as compared to normal liver tissue. And also we used nature compounds that inhibited lysyl oxidae. The result of this study suggested that lysyl oxidase maybe required for normal liver region to become cirrhotic. Our findings indicate that to inhibit LOX maybe a good therapeutic target to prevent tumor growth.
