橄欖果萃取物對於高脂血症倉鼠之脂質代謝、抗氧化活性與粒線體功能的影響

Effects of olive fruit extract on lipid metabolism, antioxidant activity and mitochondrial function in hyperlipidemic hamsters

摘要


高脂血症為體內脂質代謝異常之疾病,易演變成動脈粥狀硬化,為心血管疾病的危險因子。橄欖果中含有許多酚類化合物,其中hydroxytyrosol 為主要酚類化合物,具有抗氧化、改善粒線體功能及降低血中脂質之功效。本實驗藉由高脂高膽固醇飲食誘發倉鼠高脂血症,給予橄欖果萃取物後,能否改善脂質代謝異常、抗氧化酵素活性及胰島素抗性,並評估對粒線體功能可能影響。本實驗使用5週齡雄性倉鼠,適應兩週後依體重隨機分為六組,控制組 (C);高脂高膽固醇飲食組 (N);高脂高膽固醇飲食給予低劑量 (L)、中劑量組 (M)、高劑量橄欖果萃取物 (H) 及正控制組為高脂高膽固醇飲食給予N-Acetyl Cysteine (NAC) (P),介入八週後予以犧牲,取其血液、肝臟、脂肪及糞便進行分析。實驗八週後,發現給予橄欖果萃取物可降低血中脂質、血糖、胰島素及胰島素抗性、脂質過氧化終產物及改善粒線體質子滲漏的情形,也可改善高密度脂蛋白膽固醇、脂質氧化酵素活性、抗氧化酵素活性、粒線體功能及增加糞便中性固醇濃度。總言之,橄欖果萃取物可藉由降低因高脂高膽固醇飲食所誘發之氧化傷害,增加抗氧化酵素的活性,進而延緩粒線體功能受損,改善胰島素抗性,降低脂質過氧化,亦可增加體內膽固醇的排出,減少體內脂質的累積,達到改善血脂異常的作用。

Hyperlipidemia resulting from lipid metabolic dysfunction is a major cause of atherosclerosis and cardiovascular disease. Olive fruit has been reported that can reduce oxidative stress, anti-inflammatory, anti-cancer, and anti-hypertention due to olive polyphenols. Hydroxytyrosol (HT), a major phenolic compound in olive, has increased attention for its antioxidative activity and mitochondrial function regulation effect. The aim of this study was to investigate whether the olive fruit extract supplement attenuated high fat high cholesterol diet induced hyperlipidemia, oxidative stress, mitochondrial dysfunction and insulin resistance in hamsters. Male hamsters were randomly divided into six groups : Control group (C) : AIN-93M diet. Negative group (N) : AIN-93M modified high fat high cholesterol diet. Low dosage group : 0.04 % diet olive fruit extract with high fat high cholesterol diet. Medium dosage group (M) : 0.11 % diet olive fruit extract with high fat high cholesterol diet. High dosage group (H) : 0.18 % diet olive fruit extract with high fat high cholesterol diet. Positive group (P) : 0.14 % diet N-acetyl cysteine (NAC) with high fat high cholesterol diet. The plasma lipid levels, insulin resistance, liver antioxidant enzymes activities, liver lipid metabolism enzymes activities, mitochondrial function and stool neutral steroid were examined. After 8 weeks, in olive fruit extract groups were significantly decreased the plasma lipid and insulin resistance compared with N group. In liver lipid metabolism, in olive fruit extract groups were significantly increased lipid oxidation related enzymes activities, cholesterol metabolism related proteins expression, stool neutral steroid concentration and lowered liver lipid accumulation compared with N group. In antioxidant function, in olive fruit extract groups were significantly increased the antioxidant enzymes activity and reduced the malondialdehyde (MDA) concentration compared with N group. In mitochondrial function, in olive fruit extract groups were significantly increased the mitochondrial enzyme activity and the mitochondrial respiratory control ratio (RCR) compared with N group. These results suggested that the hypolipidemic effect of olive fruit extract might be due to their abilities to enhance antioxidant enzymes activities, slowing the lipid peroxidation process, improving mitochondrial function, lowering insulin resistance as well as lowering serum lipid.
