本研究探討豆科 ( Fabaceae ) 植物蘆巴子Trigonella foenum- graecum L.甲醇萃取物之成分。依初期之探討其乙酸乙酯層對降血糖有明顯的活性,經由各種層析管柱及不同溶媒系統層析後,共分離出十二個化合物。其結構根據物理數據、核磁共振光譜判定後,分別確定為:Uracil (1)、Uridine (2)、Adenosine (3)、Myo-inositol (4)、Mannitol (5)、2-ethoxy-6-(hydroxymethyl)tetrahydro-2H-pyran-3, 4, 5-triol (6)、Betulin (7)、Lupeol (8)、(25S)-5β-spirostan-3α-hydroxy-3-O-α- arabinopyranosyl (1→6)-β-glucopyranoside (9)、(25S)-5β-spirost-5- en-3β-hydroxy-3-O-β-glucopyranoside (1→2)-β- glucopyranoside (10)、Isovitexin (11)、Vicenin Ⅱ (12) 。 根據這些化合物骨架的不同,可將其歸類為皂素類9, 10、三萜類7, 8、黃酮類11, 12、嘧啶類1, 2、嘌呤類3、其他類4, 5, 6等。經初步之探討於10 mM濃度下1 (141%)、4 (150%)、7 (135%)、8 (157%)、11 (150%)等化合物有促進AMPK磷酸化之反應。另外,在對glucose uptake方面,發現3 (123.8%)、8 (122.4%)、11 (125.9%)、12 (125.2%)於2.6 mg/mL濃度下有促進glucose uptake的作用。
This study searched Fabaceae of Trigonella foenum-graecum L. methanol extracts of the ingredients. In the initial stage of its ethyl acetate layer on a clear hypoglycemic activity, through all kinds of different solvents and chromatography system, a total of 12 compounds isolated. Its structure in accordance with physical data, NMR, were identified as: Uracil (1), Uridine (2), Adenosine (3), Myo- inositol (4), Mannitol (5) ,2-ethoxy-6-( hydroxymethyl) tetrahydro-2H- pyran-3, 4, 5-triol (6), Betulin (7), Lupeol (8), (25S) -5 β-spirostan-3α- hydroxy-3-O-α- arabinopyranosyl (1 → 6) - β-glucopyranoside (9), (25S) -5 β-spirost - 5-en-3 β-hydroxy-3-O-β-glucopyranoside (1 → 2) -β- glucopyranoside (10), Isovitexin (11), Vicenin Ⅱ (12). Under these skeletons of different compounds, can be classified as Class saponin: 9,10;triterpenes:7, 8;flavonoids:11, 12;pyrimidine:1, 2; purine:3;alcohols:4, 5 , 6, and so on. 1 (141%), 4 (150%), 7 (135%),8 (157%),11 (150%) in 10 mM concentration of active AMPK phosphorylation reaction and 3 (123.8%), 8 (122.4%),11 (125.9%), 12 (125.2%)in the 2.6 mg / mL concentration of a significant role in glucose uptake.
