金剛纂化學成分及生物活性之研究

中 文 摘 要 金剛纂(大戟科)為常綠小喬木,原產於印度、斯里蘭卡,台灣各地則是零星的栽培。全株含多量白色乳汁,莖葉分泌之大戟膠,可供藥用。 在化學成分研究方面,本研究從金剛纂乙醇粗萃物中分離純化得到29個化合物,經由圖譜數據分析,其中有二十四個三萜類,一個黃酮類以及四個固醇類化合物。三萜類中包含 Friedelane type: 3b-friedelanol (1), 3a-friedelanol (2), 3b-acetoxy friedelane (3), friedelin (4);glutinane type: glutinone (5), 3b-glutinol (6), 3b-acetoxy-glutinane (7) ;lupane type: lupenone (8) ;taraxerane type: taraxer-14-en-3a-ol (9), 3a-acetoxy taraxer-14-ene (10), 3b-acetoxy taraxer-14-ene (11) ; oleanane type: b-amyrin (12), b-amyrin acetate (13) ;dammarane type: dammarendiol-II monoacetate (14) ; ocotillol-II acetate (15) ;simiarenane type: 3b-simiarenol (16), simiarenone (17) ;cycloartane type: cycloartanol (18), 3b-cycloart-25-ene-24-one (19), cycloart-23-en-3b, 25-diol (20), a mixture of 24-methylene-cycloartanol (21) and 3b-lupenol (22) (lupane type) ;cycloeucalane type: cycloeucalenol (23), 31-norcycloartanol (24) 。黃酮類化合物為kaempferol-3- O-rhamnoside (25) 。四個固醇類化合物為 a mixture of b-sitosterol (26) and stigmasterol (27), and a mixture of b-sitosteryl-3-O-b-D-glucoside (28) and b-stigmasteryl-3-O-b-D-glucoside (29). 在所有已分離純化物中,化合物1, 2, 3以及9具有明顯抗病毒活性,在 5 mg/ml之下其抑制率分別為 132.43, 80.94, 109.01 以及 111.01%

Abstract Euphorbia neriifolia (Euphorbiaceae) is a small evergreen tree. It originate from India and Sri Lanka, and cultivates in Taiwan. The plant has latex, which have been used as a folk medicine for dermatitis. In the current study, 29 compounds were isolated from the ethanolic extract of E. neriifolia. These compounds contain 24 triterpenes, one flavonol and 4 steroids. The triterpenes include friedelane type: 3b-friedelanol (1), 3a-friedelanol (2), 3b-acetoxy friedelane (3), friedelin (4);glutinane type: glutinone (5), 3b-glutinol (6), 3b-acetoxy glutinane (7);lupane type: lupenone (8);taraxenane type: taraxer-14-en-3a-ol (9), 3a-acetoxy taraxer-14-ene(10), 3b-acetoxy taraxer-14-ene (11) ;oleanane type: b-amyrin (12), b-amyrin acetate (13);dammarane type: dammarendiol-II monoacetat (14);ocotillone type: ocotillol-II acetate (15);simiarenane type: 3b-simiarenol (16), simiarenone (17);cycloartane type: cycloartanol (18), 3b-cycloart-25-ene-24-one (19), cycloart-23-en-3b,25-diol (20), a mixture of 24-methylene-cycloartanol (21) and 3b-lupenol (22) (lupane type);cycloeucalane type: cycloeucalenol (23), 31-norcycloartanol (24). The flavonol is kaempferol-3-O-rhamnoside (25).The sterols are a mixture of b-sitosterol (26) and stigmasterol (27), and a mixture of b-sitosteryl-3-O-b-D-glucoside (28) and b-stigmasteryl-3-O-b-D- glucoside (29). The structures of the isolated compounds were elucidated by spectral data. In addition, compounds 1, 2, 3 and 9 showed significant anti-virus activity. [Inhibition (%) (5 mg/ml): 132.43, 80.94, 109.01 and 111.01, respectively]
