摘要

近幾年來，台灣毒品的使用愈來愈嚴重，更蔓延到學生、演藝人員、軍警人員等社會上各個階層，毒品時常造成個人、家庭、治安、傳染等問題。此時需要一個可靠且快速的方法來進行毒品初篩試驗。現行濫用藥物的尿液檢驗通常分為兩階段進行，第一階段為初篩檢驗(screening test)檢驗方法快速、操作容易且不需經過任何的前處理就可上機檢驗。
一般採用免疫學分析法(immune assay)，經初篩檢驗後，若為陽性或有疑問時，需進行第二階段之尿液檢驗，以免產生誤判。第二階段確認檢驗(confirmatory test)對初篩檢驗所檢驗出為陽性的某特定之藥物或代謝物進行檢驗。為提高檢驗之可信度及準確度，目前確認檢驗方法多為氣相層析質譜儀(gas chromatography/mass spectrometry, GC/MS)，也是大多數檢驗機構所使用的分析方式。本研究文章是利用Marquis reagent呈色法對尿中的安非他命進行定性分析，以Marquis reagent呈色法與尿液安非他命反應產生橘色到棕色呈色反應，之後再將反應過後的尿液，以分光光度計使用固定波長進行掃描。結果顯示Marquis reagent呈色法適合實驗條件為室溫(25℃-27℃) 、非日照以及短時。由實驗結果可以發現Marquis reagent呈色法比起一般我國現行免疫學分析法來的快以及便宜，在日後發展上或許可以取代現行公告方法，提供毒品篩檢一個更快速更有效率的方法。

In recent years, the abuse of drugs in Taiwan has become more and more serious, but also spreads to students, entertainers, military and police personnel and other social strata. Drugs often cause personal, family, law and order, infection and other issues. A reliable and rapid method is needed to carry out the drug screening test. Urine testing of currently abused drugs is usually divided into two phases. The first stage is a screening test. It is a quick test method, easy to operate and can be tested without any pre-treatment. Immunological analysis (immune assay) is generally used, after a screening test showing positive or doubt, for the second phase of the urine test to avoid misjudgment. The second phase called confirmatory test is designed for a specific drug or metabolite tested as a positive test by a screening test. In order to improve the reliability and accuracy of the test. The current confirmatory test is the method of gas chromatography mass spectrometer (GC/MS). It is widely used by most inspection agencies. In this study, Marquis reagent was used to qualitatively analyze amphetamine in urine. After the Marquis reagent was reacted with the amphetamine and showed orange to brown color, the urine after the reaction was scanned with a fixed wavelength by using a spectrophotometer. The analytical results were explored and discussed. The results show that the Marquis reagent is suitable for the experimental conditions for room temperature (25°C to 27°C), non-sunshine and short time. It can be found from the experimental results that the Marquis reagent is faster and cheaper than the general immunological analysis in China. It may be possible to replace the existing announcement method in the future and provide a faster and more efficient method for drug screening.
