以延伸式閘極感測場效電晶體研製中草藥之小檗鹼感測器

Design the Berberine Sensor of Chinese Herbal Medicine Using the Extended Gate Field Effect Transistor

摘要


有鑑於中藥之副作用小,近幾年來於臨床上廣泛之研究與應用逐漸受到國際間的重視與肯定。小檗鹼(Berberine)係衍生自中藥黃連,其具有許多藥理作用,例如抑菌、抗黴菌、治療高血壓、高血脂、心律不整、糖尿病與抗血小板凝集等。近年來亦有許多研究發現小檗鹼具有治療癌細胞之功效,故小檗鹼之研究於藥理學上具有相當之重要性。 本研究係以SnO2/ITO玻璃作為酸鹼感測器,以偵測中藥材中之小檗鹼(Berberine),此元件具有全固態化、體積小、成本低且製程容易等優點。此研究主要包含二種方法,第一種方法係固定乙醯膽鹼脂酶(Acetylcholinesterase; AchE)形成酵素感測場效電晶體(Enzyme Field Effect Transistor;EnFET);第二種方法為酸鹼感測器以聚氯乙烯(Polyvinylchloride; PVC)包埋矽鎢酸(Silicotungstic acid; H4O40SiW12)形成聚氯乙烯膜感測場效電晶體(Poly vinyl chloride membrane Field Effect Transistor;PVC membrane FET)。以矽鎢酸為電活性物質之聚氯乙烯感測場效電晶體偵測線性範圍約介於1×10-3~5×10-7mol/L,而酵素感測場效電晶體約介於1×10-3~1×10-6mol/L,而其偵測極限為1×10-7mol/L。聚氯乙烯感測場效電晶體與酵素感測場效電晶體相較之,聚氯乙烯感測場效電晶體具較低之成本、較佳感測度與反應時間較短,且製程簡單。故聚氯乙烯感測場效電晶體為較佳之小檗鹼感測器。

Traditional Chinese medicine attaches importance to research in international development and in recent years concentrated dosage form have been widely adopted for clinical use, which have few side effects. Berberine is an alkaloid that is derived from Coptis chinensis. It has a lot of pharmaceutical efficacy such as antibacterical, antifungal, treated hypertension, hyperlipemia, arrhythmia, diabetes mellitus and antiplatelet effect, etc. Recently, many literatures reported to process anti-tumor promoting activities effects so berberine attaches importance application at the pharmacology. In this study, the SnO2/ITO glass as a pH sensor is used to detect berberine (BE) for traditional Chinese medicine. It is a solid-state sensor, which has a lot of advantages, such as small size, low cost and easy manufacture. The research includes two methods. First, acetylcholinesterase (AchE) is immobilized on the substrate to fabricate an enzyme field effect transistor (EnFET). Secondly, the pH sensor, which is modified with silicotungstic acid (STA; H4O40SiW12) and polyvinylchloride (PVC) is presented the poly vinyl chloride membrane field effect transistor (PVC membrane FET). The PVC membrane FET have detection linear range 1×10-3~ 5×10-7mol/L; the detection linear range of EnFET is 1×10-3~ 1×10-6mol/L and detection limit about 1×10-7mol/L. Compare PVC membrane FET with EnFET, PVC membrane FET has lower cost, better sensitivity, shorter response time and easier manufacture. So PVC membrane FET is better choice for berberine sensor than EnFET.
