南瓜萃取物之抗氧化活性與其對高脂飲食及STZ誘導糖尿病小鼠之降血糖功能分析

摘要


南瓜屬於葫蘆科南瓜屬一年生草本蔓性植物,是世界著名且分佈相當廣泛之食材,夏秋季節瓜菜之一。本研究在探討中國阿成及特殊品種兩種南瓜樣品(粉末)代測物,進行抗氧化活性比較及對於高脂飲食合併STZ誘導第二型糖尿病小鼠血糖濃度的影響。抗氧化能力測試分別做了清除DPPH自由基、螯合亞鐵離子、還原力、總多酚、總黃酮測試及總抗氧化能力和利用南瓜水、乙醇萃取物餵食小鼠,將小鼠分為六組:正常組、糖尿病組、餵食0.5 mg/kg Glibenclamide及餵食代測物劑量為10、50、100 mg/kg為期28天比較其降血糖功效以及評估糖尿病小鼠肝指數、膽固醇、三酸甘油脂和每週飲食、飲水量。結果顯示,抗氧化能力依賴濃度劑量提升,EECMX在清除DPPH自由基能力方面清除率高達85.78±1.37 %;還原程度達96.27±0.76 %;總酚、總黃酮含量含有233.33±9.18 mg of GAE/g、22.41±2.21 mg of Quercetin/g;總抗氧化能力達83.32±0.94 %;WECMX有相當好的螯合效果,清除率為77.96±0.00%。 糖尿病試驗中,餵食WECMX 100 mg/kg樣品禁食血糖值從214.75 mg/dl經過四週治療血糖平均降至159.25 mg/dl;同時,也少部分的降低了肝脂數、總膽固醇及三酸甘油脂。抗氧化結果得知南瓜是一種極有潛力的天然抗氧化劑,在動物實驗中並能夠有效控制第二型糖尿病小鼠血糖。

Pumpkins belong to the family of Cucurbitaceae. It is world famous and a wide range of ingredients in summer season. Hypoglycaemic and antioxidant effects of Pumpkin extract on high-fat diet streptozotocin-induced diabetic mice in this study. The analysis antioxidative activity include DPPH radical scavenging activity, ferrous ions chelating effects, reducing power activity, total polyphenols, total flavonoids and trolox equivalent antioxidant activity. The C. moschata extracts also was used to feed diabetic mices for the analysis hypoglycemic effects. The animals were divided into six groups, respectively normal control group, diabetic control group, diabetic fed 0.5 mg/kg Glibenclamide and oral administration of C. moschata (10,50,100 mg/kg) to high-fat diet fed doses of STZ induced type 2 diabetic mices for 28 days, and diabetic mice liver index measured, cholesterol, triglycerides and weekly diet, water intake. The results showed that the concentration of the dose-dependent antioxidative activity increase, EECMX effect of the clearance rate 85.78±1.37 % in DPPH radical scavenging activity; reducing power level up to 96.27±0.76 %;WECMX concentration 100 mg/ml have a fairly good chelating effect, clearance rate 77.96±0.00 %; total phenol and total flavonoids containing 233.33±9.18 mg of GAE/g, 22.41±2.21 mg of Quercetin/g, trolox equivalent antioxidant activity 83.32±0.94 % . Diabetes tests, feeding 100 mg/kg WECMX fasting blood glucose level from 214.75 mg/dl dropped to 159.25 mg/dl after four weeks. However, a small reduction in the number of liver lipids, total cholesterol and triglycerides. Antioxidant result that pumpkin is an extremely promising natural antioxidants in vitro, and can effectively control blood glucose in diabetes mices.
