摘要


肺癌是世界上導致死亡的主要癌症之一,這被相信是由於缺乏有效的癌篩檢和治療方法造成的,因此對於了解肺癌生物標記(biomarkers)及分子間機制以改善肺癌的檢測和治療顯得特別重要。在過去我們證明了幾個肺癌生物標記,那些標記皆顯示對於肺癌細胞的生長與增殖(proliferation)的調控是很重要的,目前的研究我們則去證明下游基因CASP-4和IGFBP-6會因為那些生物標記而受到調控,我們利用資料採礦(in silico data mining)方式尋找與過去調控基因最可能的下游基因表現圖譜(expression profiles)並透過實驗分析加以確認,從細胞培養實驗中我們也顯示那些下游基因在肺癌細胞中具活性功能,且有機會發展成為肺癌治療標靶的潛力,我們的研究提供了在肺癌中具重要的訊息傳導之候選基因。

Lung cancer is the leading cause of cancer related death in the world, which is believed to be caused by lack of effective lung cancer screening and treatment methods. It is therefore important to identify lung cancer biomarkers and to understand their underlying cancer biology/mechanisms to improve lung cancer detection and therapy. We have previously identified several lung cancer biomarkers of lung carcinoma. Those markers have been shown to be important regulators required for growth and proliferation of lung cancer cells. In the present study, we show that CASP-4 and IGFBP-6 could be downstream genes regulated by those biomarkers. We performed in silico data mining and validation assays to demonstrate that the expression profiles of downstream genes were significantly associated with our previously identified regulators. Via cell culture experiments we also showed the some of those downstream genes were indeed functionally active in lung cancer cells and are potential to be developed as target of lung cancer treatment. Our study has provided novel candidate members of the important signaling pathway of lung cancer.
