灌食不同劑量與製程的兒茶素類物質對小鼠抗氧化能力與肝功能之影響

Effects of Enteral Feeding of Catechins at Various Doses Using Different Preparation Methods on Antioxidative Activity and Liver Function in Mice

本實驗的主要目的是在探討不同劑量及製程的兒茶素類物質,對小鼠之抗氧化與肝功能的影響;將30隻雄性小成鼠隨機分成五組,各組每天皆灌食一次;其中一組灌食的配方中不含兒茶素類物質,作為控制組(CK);其他組別的處理分別依據每公斤小鼠體重,灌食本實驗室自製兒茶素類物質0.5(低劑量組,LDM)或50(高劑量組,HDM)毫克,以及大陸產製之兒茶素類物質0.5(低劑量組,LDP)或50(高劑量組,HDP)毫克。實驗期間因為HDP組比其他各組有較高的致死效應,故於第六天起改為控制組條件繼續試驗(HDP→CK)。三週實驗期完成後,結果顯示HDM組顯著提升肝臟與血清總抗氧化活性(TEAC)及降低血球脂質過氧化值(TBARS),但亦比它組有顯著偏低的最終體重與餵食效率。LDP組除了比LDM組有較高肝中AST和ALT值外,對於小鼠各項測試值都相當相似。綜合實驗數據,攝取兒茶素類物質應可增加肝及血清總抗氧化活性及血球之抗脂質過氧化能力,然其生理效應明顯又受到兒茶素類物質之劑量或製程的影響。

The purpose of this experiment was to investigate the influences of various doses and preparation methods of catechins from tea on antioxidative activity and liver function in mice. Thirty adult male mice were divided into five groups, and all were fed enterally every day. One group did not have catechins in the enteral feeding formula and served as the control group (CK). The others were administrated 0.5 or 50 mg/kg catechins made in our laboratory (LDM for low dose, and HDM for high dose) and a product from mainland China (LDP for low dose, and HDP for high dose) in the enteral feeding formula, respectively. However, the experimental condition of the HDP was changed to CK (HDP→CK) on the sixth day because of a high lethal effect. After 3 weeks of feeding, the HDM group showed a lower final body weight and feed efficiency, but the total antioxidative activity (TEAC) of the liver and serum had increased, and the thiobarituric acid-reactive substance (TBARS) value of blood cells had significantly decreased. The LDM and LDP regimens had similar effects on mice except that the LDP regimen induced higher hepatic aspartate transaminase (AST) and alanine transaminase (ALT) activities than did the LDM regimen. The results of this study indicate that catechins may be helpful in elevating the total antioxidative activity of the liver and serum in mice, and preventing the peroxidation of lipid in blood cells, but these physiological effects may also be significantly affected by dosage and preparation methods of the catechins.
