臺灣紫芝水萃取物對常見致病菌的抑菌潛力探討

Evaluation of Antimicrobial Effect of Ganoderma formosanum Water Extracts toward Common Pathogens

摘要


本研究目的係利用臺灣紫芝(Ganoderma formosanum)胞外多醣和菌絲體以3種破菌方法(液態氮、高溫高壓、研磨)之萃取物,對常見致病菌(Salmonella entericaserovar Typhimurium BCRC 10747, Escherichia coli BCRC 10239, Staphylococcus aureus BCRC 10451)進行抗菌活性測驗。結果顯示,紫芝萃取物以液態氮破菌的水層劃分抑菌效果最好,MIC分別為E. coli BCRC 10239: 2.03 ± 0.12 mg/mL; S. aureus BCRC 10451: 1.90 ± 0.1 mg/mL; S. Typhimurium BCRC 10747: 1.97 ± 0.15 mg/mL,並經生長曲線確定24小時內水層劃分萃取物都具有抑菌作用;除此之外,紫芝萃取物對三株致病菌均有殺菌效果,MBC為E. coli BCRC 10239: 2.96 ± 0.058 mg/mL, S. aureus BCRC10451: 2.80 ± 0.12 mg/mL, S. Typhimurium BCRC 10747: 2.91 ± 0.058 mg/mL;另外,在2 mg/mL濃度下顯著性抑制群體感應(抑制94.73%紫色色素)在MIC濃度下(1.90 mg/mL)也降低60.80% S. aureus的BCRC 10451生物膜合成。而在1/2 × xMIC作用下亦有抑制群體感應與生物膜合成的趨勢。

The purpose of this study is to use the extracellular polysaccharides and mycelial extracts of Ganoderma formosanumby three extraction ways (liquid nitrogen freeze-thawing, autoclave, milling), to test their antibacterial activity against common pathogens (Salmonella entericaserovar Typhimurium BCRC 10747, Escherichia coli BCRC 10239, Staphylococcus aureus BCRC 10451). The results showed that the extract of G. formosanumis best divided by the liquid layer of liquid nitrogen-cell breaking, and the MIC were E. coli BCRC 10239: 2.03 ± 0.12 mg/mL, S. aureus BCRC 10451: 1.90 ± 0.1 mg/mL, S. Typhimurium BCRC 10747: 1.97 ± 0.15 mg/mL, respectively, and all the inhibitory effects were confirmed by the 24-hour growth curves. In addition, G. formosanum also have MBC against tested pathogens (E. coli BCRC 10239: 2.96 ± 0.058 mg/ mL, S. aureus BCRC 10451: 2.80 ± 0.12 mg/mL, S. Typhimurium BCRC 10747: 2.91 ± 0.058 mg/mL). Besides, G. formosanumliquid layer significantly inhibited quorum sensing (inhibition of 94.73% purple pigment) and reduced 60.80% S. aureus BCRC 10451 biofilm synthesis at MIC concentration. There is also a tendency to inhibit quorum sensing and biofilm synthesis under the action of 1/2×MIC.
