探討五爪金英對神經膠母細胞瘤之抑癌效益

Investigating the Suppressive Effect of Tithonia Diversolia on Glioblastoma

摘要


神經膠母細胞瘤(glioblastoma,GBM)是成人常見的原發性惡性腦瘤,病人致死率極高。雖然現在對GBM治療機制愈來愈清楚,但是對GBM的標準治療並無顯著進步,幾十年來只能沿用舊有治療程序,持續研發新藥物或治療策略有迫切需求。因此本研究擬探討五爪金英葉片萃取物對GBM的抑癌效益。本研究選取有p53突變特性的U373細胞進行五爪金英萃取物的抑癌實驗,以MTT assay進行細胞的活性檢測,以及利用Annexin V/PI雙染色,評估細胞的凋亡狀態,最後再以西方墨點法測量凋亡相關蛋白質之表達量。結果顯示五爪金英葉片萃取物含有的抗癌主成分-Tagitinin C含量十分豐富,約占萃取物的44.2%±1.3%。葉片萃取物對U373標的活性抑制效果十分顯著,也能測得約32%±8%細胞凋亡,及凋亡相關蛋白質中的PUMA及caspase 8表達量增加。從這些證據可推論五爪金英葉片萃取物對GBM腫瘤細胞具有良好抑癌效益。

Glioblastoma is the most common malignant primary brain tumor in adults and is almost invariably lethal. Despite growing knowledge of the various mechanisms underlying treatment failure, the standard-of-care therapy has not changed over the last two decades, revealing a great unmet need. Herein, we investigated the antitumour activity of Tithonia diversifolia (TD) leave extract on glioblastoma cells. We examined the antiproliferative activity of TD leave extract in human p53 mutated glioblastoma cell line. Cell variability was performed using MTT assay, and apoptosis elevation using the Annexin V/PI double staining. To further test the apoptotic induction capacity of TD leave extract, the protein level of PUMA, caspase 3 and caspase 8 were determined by Western blotting analysis. The results suggested the main antitumor component was Tagitinin C abundant in TD leave extract estimated as 44.2%±1.3%. TD leave extract treatment significantly suppressed the growth of glioblastoma cells and resulted in 32%±8% glioblastoma cells proceeding apoptosis, enhancing the expression level of PUMA and caspase 8. These findings suggested that TD leaves extract was effective for confronting glioblastoma cell.
