摘要


春石斜蘭經無菌播種所得莖頂分別置於MS、花寶1號(H1)和花寶2號(H2)基本培養基中,以培養於花寶2號與花寶2號(H2)+椰子汁(CM)300 ml/l之處理,褐化率最低,類原球體(PLBs)增殖數最高。有機物添加以椰子汁對於類原球體的增殖率優於香蕉泥(Ba)及蘋果汁(Ap);添加不同濃度BA(0.5、1.0、2.0、4.0 mg/l)培養基皆可促進類原球體的形成,尤其以BA 0.5 mg/l及1.0 mg/l處理效果最佳,每處理平均產生36.9個及19.1個類原球體,褐化率卻高於內含椰子汁處理者。不同培養方式中以MS+BA 0.9 mg/l固體優於液體振盪培養,褐化率亦較低。對類原球體(PLBs)進行不切割(0刀)及切割1、2刀試驗中,顯示PLBs增殖率以不切割的處理較佳。發根試驗以花寶1號(H1)+蘋果汁(Ap)100 ml/l根數最多,添加香蕉泥(Ba)雖可誘導根之形成,但植株生長情形卻不佳。添加不同種類生長素處理以花寶2號(H2)+IBA 0.5 mg/l和花寶2號(H2)+NAA 2.0 mg/l均具良好發根情形。

Shoot tips obtained from of Dendrobium nobile in vitro were cultured in various basal medium, MS, Hyponex NO.1(H1)and Hyponex NO.2(H2). Protocorm-like bodies formation was observed to be enhanced and tissue browning rate decreased by H2 or H2(H2)+ 30 % CM(coconut milk), and CM was better than the other organic compounds such as apple juice and banana extract in cultured medium. All the concentration of BA(0.5, 1.0, 2.0 and 4.0 mg/l)treatment were beneficial for PLBs formation and 0.5 mg/l, 1.0 mg/l were best concentration for PLBs formation, but it was observed higher browning rate than those cultured in CM medium. Cutting protocorm once, twice and control showed that PLBs without cutting and cultured in MS +Ba 0.9 mg/l solid medium gave the lowest browning rate and highest shoot regeneration which is better than those cultured in shaking liquid medium. Root induction experiment showed Hyponex NO.1(H1) + Apple juice(Ap)100 ml/l treatment gaved the highest root number. Addition of banana extract(Ba) although enhanced root formation shoot development was poor. Addition of mineral and plants hormone showed both H2 + IBA 0.5 mg/l and H2+NAA 2.0 mg/l gave good root development.
