Culturing Peanut (Arachis hypogaea L.) Hairy Roots on A Simplified Medium and Evaluating Their Biomass and Trans-resveratrol Production

摘要


本研究為求降低培養耗材之花費,使用花寶1 號肥料(Hyponex No.1) (2.0 g L^(-1))混合馬鈴薯(Solanum tuberosum L.) 抽出物(potato extract)製成簡化培養基(simplified medium,代號H1-2PE),以培養落花生(Arachishypogaea L.)毛狀根(hairy root)生產反式白藜蘆醇(trans-resveratrol; t-Res)。毛狀根在H1-2PE 培養基中連續繼代4 次(180 天)後,與培養在對照組MS 培養基中者相比,其鮮重增加及t-Res 含量均未顯著減少,且生長亦未衰退。H1-2PE 培養之毛狀根在誘引因子幾丁聚醣(chitosan)處理後,可增加t-Res含量約2 倍(達6.14 μg g^(-1) FW)。以紫外光(ultraviolet light; UV)照射則增加約4.5 倍(達15.78 μgg^(-1) FW)。以配製50 L 培養基作為比較基礎, H1-2PE 培養基成本為MS medium (速溶粉末,Sigma)的8%,應具備於大規模培養時之應用價值。

A low-cost simplified medium (H1-2PE) was applied to culture peanut (Arachishypogaea L.) hairy roots for trans-resveratral (t-Res) production. The medium included Hyponex No. 1 water-soluble fertilizer and potato (Solanum tuberosum L.) extract as major components. The fresh weight increased and t-Res content of the hairy roots grown on H1-2PE medium for 4 subcultures (180 days) were not significantly different from those incubated on MS medium (control). When elicitors were introduced into the culture system, hairy roots incubated on H1-2PE medium were elicited to increase t-Res amount by 2 times (6.14 μg g^(-1) FW) with chitosan stimulation or 4.5 times (15.78 μg g^(-1) FW) with ultraviolet light illumination, respectively. To prepare 50-L culture medium, the cost of the formula H1-2PE was only 8% of those of the MS medium (dried powder, Sigma), which indicates an economical potential of H1-2PE formula in alarge-scale cultivation.
