石斛萃取物對於預防白內障效果之評估

Protective effects assessment of Dendrobium officinale extract against UV-induced zebrafish cataract

摘要


幾個世紀以來,白內障一直是損害視力的全球性問題。據估計,在未來20年內,白內障患者的數量將增加約三分之一,並且白內障仍將是世界所有地區視力損害的主要原因。產生白內障的原因為水晶體內蛋白質不正常堆疊導致水晶體呈現不透明,讓水晶體失去屈光的功能,進而影響視力。太陽光中的紫外線是造成白內障產生的因素之一,水晶體在照射紫外線會產生自由基,如果產生的自由基超過了水晶體本身的修復機制能復原的範圍,將會導致蛋白質開始朝向不正常堆疊的趨勢發展。先前研究發現,鐵皮石斛具有各種生物活性,其中包括了抗氧化能力,而白內障屬於一種氧化損傷,鐵皮石斛其中一個成分為柚皮素,此成分也具有抗氧化的效果。因此本實驗觀察餵食鐵皮石斛萃取物的斑馬魚對於延緩白內障發生的效果。先使用成年純種斑馬魚各組別15隻,每隻一天餵食一次各種萃取物(蒸餾水、酒精萃取物、水萃取物、柚皮素)餵食7天,7天後每天照射UVC在每條魚的右眼325 mJ/cm2,每天照射一次持續14天或是出現成熟期白內障即停止。統計的數據顯示,餵食酒精萃取物和柚皮素相較於餵食水萃取物和蒸餾水的組別有延長發生中度和重度白內障的趨勢,進行了水晶體內的SOD的活性分析,也顯示出酒精萃取物和柚皮素會使斑馬魚水晶體內的SOD活性增加。本研究結果未來或許能提供一個依據讓後續的實驗進一步分析鐵皮石斛哪些萃取物能讓水晶體抗氧化能力提升,達成延緩白內障發生的能力。

For centuries, cataracts have been a global problem that damages vision. It is estimated that in the next 20 years, the number of cataract patients will increase by about one-third, and cataracts will continue to be the main cause of visual impairment in all regions of the world. Cataracts are caused by the abnormal stacking of proteins in the crystal body, which causes the lens to appear opaque, which makes the lens lose its refractive function and affects vision. The ultraviolet rays in the sun are one of the factors that cause cataracts. The lens will generate free radicals when irradiated with ultraviolet rays. If the generated free radicals exceed the range that the repair mechanism of the lens itself can recover, it will cause the protein to begin to aggregate abnormally. Trend development. Previous studies have found that Dendrobium officinale has various biological activities, including antioxidant capacity. Cataracts are a kind of oxidative damage. One of the components of Dendrobium officinale is naringenin, which also has antioxidant effects. Therefore, this experiment observes the effect of zebrafish fed Dendrobium officinale extract on delaying the occurrence of cataracts. First, 15 adult purebred zebrafish in each group are used, and each fish is fed with extracts (distilled water, alcohol extract, water extract, naringenin) once a day for 7 days. After 7 days, each fish is irradiated with UVC every day. Its right eye was 325 mJ/cm2, and it was irradiated once a day for 14 days or stopped when mature cataract occurred. Statistical data shows that compared to the group fed with water extract and distilled water, feeding alcohol extract and naringenin has a tendency to prolong the occurrence of moderate and severe cataracts. The activity analysis of SOD in the lens also shows that alcohol extract and naringenin will increase the SOD activity in the zebrafish crystal. The results of this study may provide a basis for subsequent experiments to further analyze which extracts of Dendrobium officinale can increase the antioxidant capacity of crystals and achieve the ability to delay the occurrence of cataracts.
