人類精蟲中類泛素蛋白修飾分子作用與活性氧化物的關聯性

The Relevance Between of Ubiquitin-Like Small Molecule Protein Modification and Reactive Oxygen Species in Human Spermatozoa

摘要


研究目的:依據衛生福利部國民健康署108年人工生殖施行結果分析報告指出,在台灣參與人工生殖中的單純男性因素,所導致的不孕症占了不孕症因素的10.9%,位居第三位。本研究針對原因不明及輕微男性不孕症患者,活性氧化物及類泛素分子修飾分子(small ubiquitin-like modifiers, SUMOs),對於精蟲生成及功能損壞之角色進行研究,以釐清精蟲細胞活性氧化物強度與細胞中類泛素化程度的相關性。 研究方法及資料:本研究以中山醫學大學附設醫院生殖中心臨床所採之精液檢體進行研究分析,資料收集期間2017年10月17日至2018年8月27日,共計蒐集70個精液樣本納入本研究中。以史皮爾曼相關性分析SUMO相關蛋白以及活性氧化物質之間的相關性與顯著性;以及用曼–惠特尼U檢定SUMO相關蛋白在特定位置有表現與未表現的樣本組別間,其活性氧化物質之活性是否有顯著差異。 研究結果:本研究發現活性氧化物質的活性的確與SUMOs有相關性,NPC與SUMO1的表現與活性氧化物質之活性呈現正相關性(相關係數NPC為0.477,SUMO1為0.283)且有顯著性(P值,NPC為0.001,SUMO1為0.036)。 結論與建議:本研究或許能提供臨床上診斷不孕症參考,除了活性氧化物質的活性及SUMO1的表現位置或表現量可以在臨床診斷上提供一些幫助,在人類精蟲的研究當中SUMO1與活性氧化物質、氧化壓力之間因果關係仍有許多未解之處,需要後續研究來釐清。

Objective: According to statistics from the National Health Administration of the Ministry of Health and Welfare in 2020, one in every seven couples in Taiwan is infertile, and more than 10% of their infertility can be attributed solely to male sperm dysfunction. This study aims at patients with unexplained and mild male infertility. We studied the role reactive oxygen species (ROS) and small ubiquitin-like modifiers (SUMOs) on sperm function impairment. Materials and Methods: This study uses semen samples collected from the clinic of the Reproductive Medicine Center at Chung Shan Medical University Hospital. From October 17, 2017 to August 27, 2018, a total of 70 semen samples were collected. Spearman's correlation was used to analyze the correlation and significance between SUMOs and SUMOs-related proteins and ROS. and Mann-Whitney U test was used to compare the intensity of SUMOs of SUMO-related proteins in specific locations of spermatozoa between the semen samples separated by density gradient centrifugation (DGC) methods. Results: This study found that the activity of ROS is indeed related to SUMOs. The expression of nuclear pore complex (NPC) and SUMO1 and the activity of ROS are positively and significantly correlated (p-value NPC: r = 0.477, p value = 0.001; SUMO1: r = 0.283, p value = 0.036). Conclusions and recommendations: This study may provide an idea for the clinical etiology of male or unexplained infertility. In addition to the activity of ROS and the position or amount of SUMO1 expression, it can provide some help in clinical diagnosis and management for infertile couples.
